Objective:The author will inject the LEV into SD rats in advance and then observe the change of behavior of SD rats suffering from migraine induced by NTG so as to figure out the influence of LEV on behavior of those rats and on the interneurons of Glutamate and GABA which is in the upper cervical cord,with a view to see whether LEV can cure migraine and discussing why LEV could have those functions.Methods:1.We fed 24 healthy male SD rats for a week,and then divided them into False model group(6)and Model group(18)by using Strata randomized sampling method.The rats in the model group will be injected subcutaneously with nitroglycerin(the NTG),while the false model group will be injected with the same amount of normal saline.Then further divide the the experimental migraine rats model group randomly into three groups,that is,the model control group(6),group with low dose of intervention(6)and group with high dose of intervention(6).False model group: normal saline was given by intragastric administration,one time a day for seven days.And they will be fed as normal.After the seventh gavage they will be injected subcutaneously with normal saline.Model control group:normal saline was given by intragastric administration,one time a day for seven days.And they will be fed as normal.After the seventh gavage they will be injected subcutaneously with NTG to induce migraine.Group with low doses of intervention: Levetiracetam was given by intragastric administration by 100mg/kg,one time a day for seven days.And they will be fed as normal.After the seventh gavage they will be injected subcutaneously with NTG to induce migraine.Group with high doses of intervention: Levetiracetam was given by intragastric administration by 200mg/kg,one time a day for seven days.And they will be fed as normal.After the seventh gavage they will be injected subcutaneously with NTG to induce migraine.2.Then observe the change of behavior of rats and record the number of times per 30 minutes when rats scratch head,climb cage,and throw their heads.3.Three hours after those rats get migraine,inject anesthesia in the abdominal cavity,perfused with formaldehyde,and then take 1-2cervical cord.Next,dye glutamate and GABA through immunohistochemical method,observe them with the help of ordinary optical microscopy and to record the number of positive cells.Results:1.Behavioral changes: compared with false model group,the rest groups demonstrate more times of scratching head,climbing cage and throwing the head after the injection of NTG(P<0.05).Compared with model control group,the group with drug intervention shows the number of times of rats scratching head,climbing cage and throwing head decreases with the growth of dose of injection.2.Glutamate and GABA immune positive cells: compared with false model group,the glutamate immune positive cells in the rest groups were obviously increased(P<0.05),while the GABA immune positive cells were obviously decreased(P<0.05);Compared with model control group,the glutamate immune positive cells in the low and high dose groups were obviously decreased(P<0.05),while the GABA immune positive cells were obviously increased(P<0.05),these effects were in a dose dependent manner.Conclusion:LEV is effective in relieving the behavior and symptom in the rat model of migraine,and could reduce the appearance of glutamate,and increase the appearance of GABA.The mechanism may be related to regulating cervical spinal interneurons "excitatory—inhibitory" steady state of balance.
