Objective:To observe the effect of different doses of monascin on neurological deficit,brain water content,iron staining and PPAR-γ,Nrf2,SRA immunohistochemical staining at different time after intracerebral hemorrhage in rats.To study the possibility of the brain protective effects and mechanism of monascin after intracerebral hemorrhage in rats,and screen the optimal dose.Methods:A total of 120 Sprague Dawley rats,half male and half female,were randomly divided into 5 groups: the sham group,intracerebral hemorrhage group,low dose group,middle dose group and high dose group.And each group was divided into 4 time point: 1 day,3 days,7 days,14 day.Sprague Dawley rats received an infusion of Collagenase IV into the right caudate nucleus and evaluated neurological function score by Modified Gorcia Score in 1,3,7day.Rats were executed at correspoding time point and brain was taken out to measure the brain water content,carry out iron staining and immunohistochemical staining.Results:1.The total operative mortality of the model rats was 6.3%,and the model was successful.2.Compared with the sham group,neurological deficit score in all time point of intracerebral hemorrhage group,low dose group,middle dose group and high dose group was decreased significantly.In contrast with the intracerebral hemorrhage group,there were no signifinance between the intracerebral hemorrhage group and low dose group in the Garcia score.The score of middle and high dose group was increased.3.Compared with the sham group,the brain water content was obviously higher than that in the above three groups.In contrast with the intracerebral hemorrhage group,there was no significant difference in low dose group.The brain water content of middle dose group and high dose group was reduced at 1day,3days.4.Compared with the sham group,the cerebral iron contents increased most obviously at 3,7,14 days.In contrast with the intracerebral hemorrhage group,there were no signifinant difference between the intracerebral hemorrhage group and low dose group,and so was the middle dose group.The cerebral iron contents in high dose group was reduced apparently at 7days,14 days.5.The expression of PPAR-γ increased in the three group compared with the sham group and they reached a plateau at 3days.In contrast with the intracerebral hemorrhage group,there were no significant difference in low dose group and middle dose group.The expression of PPAR-γ intensified in high dose group compared with the ICH group at 1day,3 days,7 days.They went to the top at 3days and dropped to the level of ICH group at 14 day.6.The expression of Nrf2 increased in the three group compared with the sham group.There were no signifinance between the intracerebral hemorrhage group and low dose group in immune cells,and so was the middle dose group.The expression of Nrf2 intensified in high dose group compared with the ICH group.7.Compared with the sham group,the expression of SRA was obviously higher than that in the above three groups.In contrast with the intracerebral hemorrhage group,there was no significant difference in low dose group.The expression of SRA intensified in high dose group compared with the ICH group at 1day,3 days,7 days.And they dropped to the level of ICH group at 14 day.Conclusion:High doses of monascin may increase the expression of PPAR-γ,Nrf2,SRA after intracerebral hemorrhage,reduce the brain edema degree and iron contents,improve the neurological deficit.Monascin may play protective effects after intracerebral hemorrhage by activating PPAR-γ-Nrf2-SRA pathway.
