| Objective: This experiment was to study the expression of CXCR4 in aplastic anaemia bone marrow cell and its effect on bone marrow hematopoietic cells,which will provide a experimental data for further clear the pathogenesis of aplastic anemia.Methods: 20 cases of the initial treatment patients with aplastic anemia was selected,bone marrow smear and bone marrow biopsy were performed on the posterior superior iliac spine,the tissues were embedded in paraffin,(1)HE staining to analysis of characteristics of bone marrow pathology;(2)we tested expression position of CXCR4 in aplastic anemia bone marrow cells by immunohistochemical technique;(3)then the semi-quantitative analysis was used to test CXCR4 protein content;(4)at last,we conducted correlation analysis between CXCR4 expression and peripheral WBC,HBG,PLT in AA group.Results:(1)Bone marrow pathologic characteristics of AA group: proliferation of nucleated cell was extremely reduced,granulocyte and red blood cells in different stages were rare,lymphocytes were common,megakaryocyte was absent,which have a lot of fat cells,while the proliferation of nucleated cell were activity,granulocyte and red blood cells in different stages that could be seen,lymphocytes distributed dispersedly and the number of megakaryocyte is normal in control group.(2)We showed that the positive cells of CXCR4 in AA group were obviously higher than control group,and positive staining was mainly in the cytoplasm by immunohistochemical technique;(3)The optical density value in AA group(0.2598± 0.0507)was significantly higher than control group(0.0740 ±0.0097),t = 15.69,P < 0.001,the difference is statistically significant;(4)The data showed that the expression of CXCR4 protein in AA group were negatively correlated with blood cells,and the correlation coefficient r of white blood cells,hemoglobin,platelet were respectively 0.949,0.865,0.839,P < 0.001.Conclusions: Comparing with the control group,the expression of CXCR4 in aplastic anemia group increased significantly,and was negatively correlated with peripheral blood cells,CXCR4 may be involved in the pathogenesis of bone marrow failure in patients with AA. |
