Objective Both isoniazid(INH)and rifampicin(RIF)were first-line drugs in antituberculosis therapy recommended by World Health Organization(WHO),which were the main reason of acute drug-induced liver injury in China.In this study,we studied the protective effect of Lactobacillus casei strain Shirota(LcS)on liver damaged induced by isoniazid and rifampin and the effects on the intestinal microflora in rats.Methods 72 male SD rats were randomly divided into 6 groups including control group(CN),drugs group(DR),drugs and low-dose LcS group(DL),mid-dose LcS group(DM),high-dose LcS group(DH)and bicyclol group(DB,each n=12).DR,DL,DM,DH and DB groups were given by gavage with INH(50 mg/kg)and RFP(50 mg/kg)per day,and after two hours,DL,DM and DH groups were given by gavage with 10×108CFU/kg,20×108 CFU/kg and 40×108 CFU/kg LcS per day,and DB group with bicylol(7.5mg/kg).CN group were given by gavage with normal saline(20ml/kg)per day.The intervention persisted for 28 days.Then 1iver index,alanine aminotransferase(ALT),aspartate transaminase(AST),alkaline phosphatase(ALP),total biliary acid(TBA),total bilirubin(TBIL),directbilirubin(DBIL),indirect bilirubin(IBIL),superoxidedismutase(SOD),malondialdehyde(MDA),Lactobacillus,Bifidobacterium,Enterococcus faecalis and Escherichia coli were tested.Results The analysis oxidative stress level indicated that the content of MDA decreased and SOD increased significantly in the DL,DM,DH and DB groups.The contents of ALT and the ALP were 65.46 U/L and 322.76 U/L in the DR group,compared with which,the contents were decreased by 16.3% and 14.4% in DH group,respectively.Compared with DR group,the levels of TBA in DL,DM and DH groups were significantly decreased by26.0%,39.4%,42.1%,and the DBIL and IBIL were significantly decreased by 22.6%,37.9% and 13%,15.4% in DM and DH groups(P < 0.05).Similarly,the TBA,TBIL,DBIL and IBIL in DB group were also significantly decreased by 29.4%,19.1%,24.7%and 18.4% than DR group(P < 0.05).The contents of Bifidobacterium and Lactobacillus in the feces of DL,DM and DH groups were significantly increased by 8.70%,42.26%,70.19% and 46.17%,49.02%,64.11%(P < 0.05),respectively,however,the contents of Enterococcus faecalis and Escherichia coli were significantly decreased by 2.43%,32.07%,45.37% and 27.01%,27.46%,21.69%(P < 0.05),respectively.Conclusion Lactobacillus casei can reduce the liver injury induced by RFP and INH in rats,and improve the disorders of intestinal flora.
