| Objective To investigate the relationship between PPARγ gene Pro12 Ala and C1431 T polymorphism and gestational diabetes mellitus(GDM).Methods As a study of 260 pregnant women all from June 2015 to February 2016 in Qingdao University Hospital obstetrics clinics for delivery and hospitalization in Qingdao area Han patients,using case-control research methods to select one of the 130 cases of gestational diabetes mellitus(Mean,28.6 ± 3.1 years),the mean body mass index(24.2 ±3.1),and the other 130 healthy pregnant women as the control group,the mean age was(28.3 ± 3.1),the average age was(39.3 ± 0.6)± 3.2)years old,the mean gestational age was(39.1 ± 0.5)weeks,the mean body mass index was(24.1 ± 2.6),and the two groups of pregnant women were no hypertension,heart disease and other pregnancy complications.The levels of fasting blood glucose(FPG),fasting insulin(FINS),triglyceride(TG),total cholesterol(TC)and body mass index were measured and the natural logarithm ln(HOMA-IR)of insulin resistance index was calculated.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)was used to identify the two sites of Pro12 Ala and C1431 T of PPARγ gene.Finally,some samples were selected for sequencing.The result is correct.Results(1)The allele frequency of Ala12 allele in GDM group was 0.054,which was significantly lower than that in control group(0.092).The difference was statistically significant.The T1431 allele frequency was 0.269,which was higher than that of control group(0.246).(P<0.05).The frequency of allele frequency was χ2=3.345(P>0.05),allele frequency χ2=1.420(P>0.05),the frequency of genotype C1431 T was χ2=1.041(P>0.05),allele frequency χ2=0.181(P>0.05).There was no significant difference in the frequency distribution between the two loci(see Table 2.1.1.2.2.1).(2)GDM group PPAR gamma Pro12 Ala gene loci,genotype Pro/Pro measurements triglyceride and fasting blood glucose were 2.85+0.89,3.92+1.06,Pro/Ala measurements and Ala/Ala genotype of triglyceride and fasting blood glucose were 2.02+1.23,3.24+0.85,carrying the Ala allele and fasting triglyceride in pregnant women the blood glucose level was significantly lower than that of Ala allele in pregnant women,the difference was statistically significant;measured value index of genotype Pro/Pro cholesterol,fasting insulin and insulin resistance were 5.33+1.08,12.95+9.25,2.28+2.36,Ala and Pro/ index measurements of genotype Ala/Ala cholesterol,fasting insulin and insulin resistance respectively.5.69+3.86,13.56+2.82,2.76+0.42,there was no significant difference in the distribution of several indicators in the two genotypes of(P>0.05).The C1431 T locus in GDM group,numerical type C/C biochemical index was 2.23+0.65,5.28+1.45,3.88+1.37,12.94+3.25,2.42+2.05,the C/T+T/T value of various biochemical indexes were2.53+1.78,5.59+3.68,4.11+0.69,13.27+2.96,2.56+0.29.Each index in carriers of different genotypes in the incidence of GDM were not found to have statistically significant difference(P > 0.05).Conclusion PPARγgene Pro12 Ala polymorphism may be related to the incidence of GDM,Ala allele may be related to GDM glucose and lipid metabolism disorders;C1431T gene polymorphism and GDM incidence and related biochemical indicators no significant correlation. |
