Objective:1.To detect the changes of the serum thyroxine in respiratory distress syndrome(RDS)rats and healthy rats at different time points.2.To detect the expression of KLF2 protein and KLF2 mRNA in lung tissue of the RDS rats and healthy rats at different time points,and to observe the pathological changes about the lung tissue of Rats,then to research the change of expression of klf2 transcription factor about the lung tissue of RDS rats and the changes of thyroid hormone level,so as to explore the interaction between klf2 and serum levels of thyroxine,It is possible to provide a theoretical basis for the clinical application of Pulmonary surfaetant in the treatment of insensitive NRDS.Methods: 50 male SD rats were randomly divided into 4 groups,the oleic acid group 39 cases,divided into 3 hour,18 hours and 24 hours group with 13 rats in each group,while the other 11 rats of the control group without any treatment.RDS rats model was established by injecting 0.17mg/Kg oleic acid into the tail vein of 39 SD rats in oleic acid group,then detection of serum thyroxine by Electrochemiluminescence in 3h,18 h,24h after injection,Also detect the expression of KLF2 protein and KLF2 mRNA in lung tissue of RDS rats and Non RDS rats in the above time perioduse by Western blot and RT-PCR methods,and observe the pathological changes of lung tissue by immunohistochemistry.Results: 1.oleic acid 3H group,18 h group of serum FT4 compared with the normal control group and oleic acid 3H group T4 compared with the normal control group,with significant statistical significance(P< 0.01),The T4 data of oleic acid 18 h group compared with the normal control group,with statistical significance(P < 0.05);2.The KLF2 proteincontent in lung tissue of oleic acid 3H group was compared with that of KLF2 protein in healthy SD rats,which was statistically significant(P < 0.05);while there was no significant difference at 18 h group and 24 h group.The results indicated that the amount of KLF2 protein of lung tissue of RDS rats caused by oleic acid has decreased when compared with the normal group in 3 hours at this animal experiments.3.The KLF2 mRNA data in lung tissue of oleic acid 24 H group and healthy SD rats had significant difference(P < 0.05);while there was also no significant difference at other two time point groups.The results indicated that the amount of KLF2 mRNA of lung tissue of RDS rats caused by oleic acid compared with the normal group in 24 hours and has a tendency to increase at this animal experiments.Conclusion: 1.The thyroid hormone FT4 、 T4 levels in RDS rats has a obviously decreased tendency in 3H and 18 h,respiratory distress syndrome and serum thyroxine levels are correlated.2.The quantity of KLF2 protein and KLF2 mRNA of oleic acid induced lung tissue of RDS rats was reduced after 3 hours compared with the normal group,which can be conclude that there has been a relation between Respiratory distress syndrome and KLF2 protein.3.The expression of FT4、T4 is decreased in the RDS rat,and the protein expression of KLF2 in lung tissue is reduced in Oleic acid three hours,therefore,FT4 and T4 have a relationship with the KLF2.
