The Relationship Between Polymorphism Of ENOS,EPO,TNF-α Gene And Pregnancy-induced Hypertension Disease Of Han Group In Qinghai Province

Objective: To study the differences of e NOS G894 T,EPO T3541 G,TNF-αG308A polymorphism between patients with hypertensive disorder complicating pregnancy and normal pregnant wormen in Han nationality of Qinghai.It was detected that the content of EPO and TNF-α in the plasma.The expression of e NOS m RNA in blood was also compared.It was studyed that the association of e NOS G894 T,EPO T3541 G and TNF-alpha G308 A polymorphism with gestational hypertension disease in order to provide basic theory for the prevention of pregnancy induced hypertension in Qinghai area.Methods: It came from major hospitals in Qinghai province that peripheral blood samples and clinical data were collected about gestational hypertension patients and normal pregnant wormen in Han nationality.Genomic DNA was extracted.It were detected respectively that e NOS G894 T,EPO T3541 G and TNF-α G308 A polymorphism by using restriction fragment length polymorphism polymerase chain reaction(PCR-RFLP)and DNA sequencing.The sequencing results were compared with NCBI data to ensure that the PCR products and the enzyme-digested products were reliable and verified.It was detected that the content of EPO and TNF-α in the plasma by using enzyme-linked immunosorbent assay(ELISA).The expression of e NOS m RNA was distinguished between the PIH group and the normal group by semi-quantitative reverse transcription and polymerase chain reaction(Sq RT-PCR).Results:1.Frequency of e NOS gene with GG,GT and TT genotype were 17.4%,82.6%,0% respectively in PIH group.The control group were 0%,95.6%,4.4% respectively.It was different of genotype frequency distribution.G and T allele frequencies in PIH group(58.7%,41.3%)were different from the control group(47.8%,52.2%).It was statistically significant(P<0.05),Gene frequency of G in PIH group was higher than the control group.The PIH group vs the control group(OR=1.553,95% CI: 1.107~2.178).G allele of e NOS G894 T was a risk factor to PIH(OR=1.229,95% CI:1.048~1.441).2.The TT,TG,and GG genotype frequency of EPO T3541 G were 9.7%,74.2%,16.1% respectively in PIH group.It were 42.9%,46.4%,10.7% respectively in the control group.The difference existed(P<0.05).TT genotype frequency in PIH group was obviously lower than the control group,TG and GG mutant genotype frequency were higher than the control group.The frequency of T and G allele were 46.8.% and53.2% respectively in PIH group.It were 66.1% and 33.9% respectively in the control group.There was significantly different between the two groups(P<0.05).The frequency of T allele was lower in the PIH group than the control group.G allele was obviously higher in the PIH group than the control group.The PIH group vs the control group(OR=0.451,95% CI: 0.311~0.655).G allele of EPO T3541 G was a risk factor to PIH(OR=1.569,95% CI: 1.263~1.949).3.The GG,GA,AA genotype frequency of TNF-α gene G308 A were 18.18%,72.73%,9.09% respectively in PIH group and 42.86%,52.38%,4.76% respectively in the control group.The difference were statistically significant(P<0.05).The sum of GA and AA frequency was 81.82% in the PIH group which was obviously higher than the normal pregnancy group(57.14%).The frequency of G allele(54.55%)in PIH group was lower than the normal group(69.05%).A allele(45.45%)in PIH was higher than the control group(30.95%).The PIH group vs the control group(OR=0.538,95% CI: 0.375~0.771).A allele of TNF-α G308 A was a risk factor to PIH(OR=1.469,95% CI: 1.170~1.843).4.It was obviously higher that the EPO content of the plasma(454.113±35.097)pg/m L in the PIH group compared with the control group(396.316±50.262)pg/m L,(P<0.01).5.There was not difference between TNF-α content of the plasma in the PIH group(21.976±17.570)pg/m L and the control group(27.796±14.730)pg/m L(P>0.05).6.The expression of e NOS gene m RNA(1.007±0.155)in the PIH group was lower than the control group(1.243±0.329),(P<0.05).Conclusion:1.It were related that e NOS G894 T,EPO T3541 G,TNF-alpha G308 A polymorphism and gestational hypertension of Han nationality were put into study inQinghai province.G allele of e NOS G894 T,allele G of EPO T3541 G and A allele of TNF-α G308 A were susceptible to pregnancy induced hypertension disease.2.The expression of EPO was obviously high in the plasma as suffering PIH.3.There was no significance about the TNF-alpha content in the plasma of the PIH group.4.There was a lower expression of e NOS m RNA in the PIH blood.
