Objective: To study the influence of CD80 molecular expression in kidney tissue of fish bile poisoning rat after hAMSCs transplation.And the mechanism hAMSCs transplantation was explored on treatment of acute renal tubular necrosis(ATN)by fish bile poisoning.Methods: Use the fish gall bladder poisoning model rat of acute renal tubular necrosis preperated by gavage administration with fish bile in the early stage.Take the third,7th and14 th days after hAMSCs transplantation from caudal vein by injection.Experiment has been divided into control group(normal rats),model group(fish gall bladder poisoning rats),and transplant group(hAMSCs transplantation rats).Paraffin-embedded sections of kidney tissue were stained by immunohistochemical staining.The change of CD80 molecular expression was observed under microscope.Integrated optical density value(IOD)was semi-quantitatively calculated by image analysis.Results:(1)The level of renal cortex CD80 molecular expression has no change in control and transplant group at the third,7th and 14 th days after hAMSCs transplantation(P>0.05).The levels of CD80 molecular expression were higher at the third day than at the7 th day and 14 th day in model group(P<0.05).At the 7th day and 14 th days were no different with control group and transplant group(P>0.05).(2)At the third day and 7th day after hAMSCs transplantation,the levels of renal cortex tubular epithelial cells CD80 moleular expression were the lowest in model group(P<0.05).At the 14 th day was no different with control group(P > 0.05).The levels of CD80 moleular expression were the highest in transplant group(P<0.05).At the 7th and 14 th days were no different with control group(P>0.05).(3)The level of renal medulla tubular epithelial cells CD80 moleular expression was not observed in the control group at third,7th and 14 th days after hAMSCs transplantation(P>0.05).The levels of CD80 moleular expression were higher at the thirdday than at the 7th day and 14 th day in model group(P<0.05).The levels of CD80 moleular expression were obviously decrease at the 7th day in model group,and the 7th day higher than the 14 th day(P<0.05).At the third day after hAMSCs transplantation,model group higher than transplant group(P<0.05).The levels of CD80 moleular were little expression at the third day in transplant group,and the third day higher than control group(P<0.05).Conclusion: hAMSCs transplantation treatment acute renal tubular injury of fish bill poisioning may inhibit the level of CD80 expression in glomeruli and renal tubular epithelial cells,thereby reducing T cell activation to reduce the inflammatory response. |
