Recently,the novel drug delivery system has been paid more attention in the biomedical field.The potent drug delivery system has numerous advantages,the main one is the ability of targeted delivery with controlled release.Such a system not only improves the therapeutic efficacy,but also minimizes the serious side effects during their clinical uses.In order to find high-powered drug delivery system,many novel drug carriers of these systems have been researched and evaluated in the biomedical field.Especially,carbon nanomaterial has attracted particular attention in this regard.They offer many potential advantages,such as ultra high surface area,high drug-loading capability,effective transportation capability and enhanced cellular uptake.The two main materials of carbon nanomaterials-carbon nanotubes and graphene oxide can achieve drug delivery and drug targeted therapy by surface modification and drug loading which make it used widely in the biomedical field.In this study,we studied the preparation of modified carbon nanomaterials and the research on the targeted therapeutic efficacy of liver cancer.(1)The product was synthesized by modifying PEI or CMC,followed by sequential modification with a tracing ligand FITC and a targeting ligand GL or LA.The resulting carrier complexes were characterized by 1H-NMR、 FTIR、 UV-Vis、 TEM and Zeta potential measurements.The results show the carrier complexes were successfully modified onto the carbon nanomaterials.(2)Doxorubicin,as a anticancer model drug,was loaded onto nanomaterials by π-π stacking interaction and the vitro release behavior of DOX was monitored via UV-vis spectrometry.Release at pH = 5.8(typical of the tumor cell microenvironment)was much more rapid and reached a greater extent than release under normal physiological conditions(pH = 7.4).The pH-sensitive release indicates that carrier complexes were potential carrieres for controlled releasing of anticancer drug in tumors,and avoiding deleterious side effects in normal tissue.(3)MTT assay was carried for determining targeted therapeutic efficacy of drug delivery system and the drug carriers’ toxicity.Cell uptake assay and confocal microscopy images were also used to evaluate the targeted potential of GL/LA with SMMC-7721 cells(human liver cancer cell)and L929 cells(mouse fibroblasts cells).The result shows that DOX carbon nano composite material with DOX loaded can actively identify SMMC-7721 cells for achieving targeted therapy.
