Polysaccharides is composed of monosacchairides that have different structures by connecting with each other with glucosidic bonds which be formed by dehydration condensation.In addition,polysaccharides have many biological activities,such as antioxidant activity,hypoglycemic,hypolipidemic activity and immune activity.Changes in chemical composition content are usually considered a primary sign that Chinese drug preparations process has a substantive change,therefore,measuring the changes in chemical composition content can be used as a useful method to evaluate whether the Chinese drug preparations process has a substantial change.Based on this,in this study,taking 20 kinds of single herbs as the research object,it was studied on the influence of the process on the transfer of polysaccharides and summed up the better process.Meanwhile,the five productions of polysaccharides as the main indicator of Chinese drug preparations have been studied for theoretical analysis,exploring of relationship between the process and the polysaccharides content of Chinese drug preparations,to provide experimental basis for the optimization of hope for the future practical production process.The content of polysaccharides had been used as the evaluation index.To optimize the extraction process of 20 kinds of single herbs polysaccharide,the effects of three different influence factors were studied,such as pretreatment,solid-liquid ratio and extraction times,on polysaccharides content of 20 kinds of single herbs.The content of polysaccharides from the optimized process and original process were compared in order to explore the effect of extraction process on the content of polysaccharides.The relationship between the process and the content of polysaccharide was summarized.In addition,in order to explore the relationship between the optimization of the process and the Chinese drug preparation compound polysaccharide content,the theory of 5 Chinese drug preparations containing above-mentioned 20 kinds of single herbs and the main indicator was analyzed for the content of polysaccharides,and extraction of original process and optimization process was compared,and the contribution rate of polysaccharide content of key herbs in the whole prescription was studied.The results showed that the determination method of polysaccharide content was stable and feasible,the specificity was good,and the recovery rate was between 96.00~102.00%,which was in accordance with the relevant provisions of the Pharmacopoeia.In the study of the influencing factors on the extraction process of 20 kinds of single herbs,the effects of pretreatment,the ratio of material to liquid and extraction times on the extraction of polysaccharides content were different;the effect of the extraction of polysaccharide from 20 kinds of single herbs was as follows: pretreatment>extraction times>the ratio of material to liquid.At the same time,it could be found that the optimization extraction of crude polysaccharide from root and rhizome could be improved obviously.In addition,the percentage of crude polysaccharide extraction was related to the properties of crude drugs;In the extraction process,the greater the extraction area and the more lose the surface,the transfer of crude polysaccharide to the extraction solvent was more conducive.In the study of the optimization process of 5 Chinese drug preparations extraction contents of polysaccharides,the extraction process was optimized and the polysaccharide content increased respectively: preparation A: 89.13%;preparation B: 92.22%;preparation C: 136.43%;preparation D: 68.56%;preparation E: 34.43%.It could be seen that the optimization of the extractive method of polysaccharide could effectively improve the polysaccharide content of whole preparation.So then,it could be verified that optimizing the extraction process on 20 kinds of single herbs could improve the content of polysaccharides.And this conclusion also could be used in Chinese drug preparations,which could guide the actual production of enterprises.In summary,this experiment through the optimization of 20 kinds of single herbsextraction process,summed up the influence trend of process on 20 kinds of single herbs,put forward the advisement of the process optimization.At the same time,by using 5 Chinese drug preparations to verify the optimization process,it was concluded that the optimization of the extraction process could be used to improve the index marker such as polysaccharides,which had a guiding significance for the actual production of enterprises. |
