Objective: long non-coding RNA H19(lnc RNA-H19),which can be transcribed into micro RNA mi R-675-5p,is reported to be a tumour suppressor in many malignant tumours.However,the regulation of cell proliferation,invasion,migration and apoptosis in tongue squamous cell carcinoma(TSCC)by lnc RNA H19 is currently not well understood.Our purpose is to determine whether lnc RNA H19 protects against the occurrence of TSCC in vivo.Materials and Methods: A total of 27 TSCC samples were obtained to detect the expression of lnc RNA H19.Then we overexpressed lnc RNA H19 in CAL27 and SCC9 cells,QPCR assay was used to confirm the changes in m RNA expression,the MTT assay was used to check the cell proliferation ability in the two cell lines,and apoptosis and cell cycle analyses were performed via flow cytometry.Cell invasion and migration ability were detected by transwell assay.Dual-luciferase reporter gene assay was used to reveal the relationship between mi R-675-5p and GRP55.Results: lnc RNA H19 was expressed at a low level in TSCC compared with the normal tissues,which verified that lnc RNA H19 might serve as a tumour suppressor in TSCC.By overexpressing lnc RNA H19,mi R-675-5p expression level was also up regulated,and leads to reduced proliferation,invasion and migration and enhanced apoptosis in CAL27 and SCC9 cells.Flow cytometry assay showed cell cycle was arrested in GO/G1.Dual-luciferase reporter gene assay revealed that mi R-675-5p was binding to GRP55 and may be a possible explanation for the changes after overexpression of lnc RNA H19.Conclusions: lnc RNA H19 inhibit cell proliferation,invasion and migration and enhance apoptosis in tongue squamous cell carcinoma,and it may function throughmi R-675-5p/GRP55 aixs.
