| Objective: To discuss the effect of bioactive peptide KLD-2 / rh BMP-2 gel on intervertebral fusion,and to find a new method for the treatment of lumbar instability.Methods: Group: 24 New Zealand white rabbits,4-6 weeks old,weight 2.0-2.5kg,male and female,were randomly divided into A,B,C group,each group of 8.Group A was experimental group: KLD-12 polypeptide / rh BMP-2 group;group B was positive control group(group rh BMP-2);group C was negative control group(KLD-12).Annulus puncture nucleus aspiration,curettage of lumbar instability model,KLD-12 / rh BMP-2,rh BMP-2,KLD-12 polypeptide polypeptide of 100 L were implanted into intervertebral disc transplantation after 2 weeks,4 weeks,8 weeks and 12 weeks of Micro-CT and X-ray,observe whether bone,trabecular bone number calculation volume,density,degree of separation,the bone volume fraction;rabbit intervertebral disc specimens collected by general observation,and HE staining.Results:(1)x ray examination: A and B two groups: there were signs of osteogenesis,the bone tended to increase at each time point;there were more irregular bone formation in B group;C group showed degeneration of vertebral body and no obvious bone formation among vertebral body.(2)CT results: 4 weeks after operation,A group of trabecular bone volume(1/mm)0.453 ± 0.062,trabecular thickness(mm)0.068 ±0.018,trabecular separation(mm)0.143 ± 0.018,bone volume fraction(%)2.274 ± 0.117;B group(trabecular bone volume0.534 ± 0.031 1/mm),trabecular thickness(mm)0.078 ± 0.009,trabecular separation(mm)0.112 ± 0.017,bone volume fraction(%)3.231 ± 0.127.After 12 weeks,A group of trabecular bone volume(1/mm)0.712 ± 0.047(mm),trabecular bone Liang Houdu 0.104± 0.022,trabecular separation(mm)0.084 ± 0.011,bone volume fraction(%)4.274 ± 0.134;B group,trabecular bone volume(1/mm)0.616 ± 0.032,bone Liang Houdu(mm)0.092 ± 0.013,trabecular separation(mm)0.097 ± 0.012,bone volume fraction(%)3.841 ± 0.131.(3)gross observation,A group had no obvious stenosis of intervertebral space between vertebral bodies,surrounded by small amounts of prominent bone formation;B group had no obvious vertebral stenosis,lumbar around more bone formation;group C intervertebral space stenosis,lumbar surrounding bone formation has obvious degeneration.(4)HE staining: 400 X and A group showed the differentiation of fibroblasts into bone cells within 8 weeks after operation.B group showed mature bone knitting at 8 weeks after operation;C group showed spindle shape in 8 weeks after operation.Conclusion: KLD-2 peptide / rhBMP-2 gel can induce interbody fusion and reduce the amount of ectopic bone formation. |
