| ObjectiveTo detect serum vascular endothelial growth factor(vascular endothelial,growth factor,VEGF),arginase 1(arginase-1,Arg-1),inducible nitric oxide synthase(Inducible nitric oxide dynthase,iNOS)concentration variation before and after microwave ablation of in patients with advanced non-small cell lung cancer(NSCLC),Analysis of the relationship between VEGF,Arg-1,iNOS three and the mechanism of tumor factor changes caused by microwave ablation therapy.MethodsThere were 28 cases of healthy human peripheral blood serum as the control group,30 cases of patients with advanced non-small cell lung cancer in peripheral blood serum as the experimental group,they were taked 10 ml venous blood before and after microwave ablation for 1 days,third days and 1 months,serum samples were stored at 2℃-8℃,in the10000 rpm under the centrifugal 10 minutes,get the supernatant frozen at-80℃,using enzyme-linked immunosorbent absorption experiment(ELISA)method for detection of healthy people,patients with advanced non-small cell lung cancer patients with serum VEGF,Arg-1,iNOS concentration.All quantitative data were expressed as mean ±standard deviation(`X±S).The two groups were compared with independent samples t test.ConclusionsThere was no significant difference in VEGF,Arg-1 and iNOS between peripheral blood serum and age,tumor histology in patients with advanced NSCLC(P value were all>0.05).The concentrations of VEGF,Arg-1 and i NOS in persons with NSCLC before microwave were 816.56±13.26 pg/ml 、 5.17±0.20 ng/ml 、 544.18±13.93 pg/ml;the concentrations in normal persons were 93.43±9.93 pg/ml、1.08±0.54 ng/ml、8.08±0.33pg/ml,t values were 239.09、110.36、210.79.The concentration of VEGF,Arg-1 and iNOS in patients with advanced NSCLC were significantly higher than those in healthy subjects,P values were 0.01、0.02、0.01,and the difference was statistically significant(P<0.05).The concentrations of VEGF in persons with NSCLC on the days of first days,third days and 1 month were 708.41±10.49 pg/ml 、 592.63±7.25 pg/ml 、 521.91±8.32 pg/ml;the concentrations of Arg-1 in persons with NSCLC on the days of first days,third days and 1month were 5.95±0.01 ng/ml、7.02±0.13 ng/ml、7.67±0.09 ng/ml;the concentrations of i NOS in persons with NSCLC on the days of first days,third days and 1 month were53.01±9.48 pg/ml 、 393.21±9.42 pg/ml 、 352.60±8.31 pg/ml.Serum VEGF before treatment was compared with that after treatment: 1 day,t = 35.05,P = 0.04,3 day,t =81.16,P = 0.01;1 month,t = 103.09,P= 0.01.After first day of treatment,the serum VEGF concentration was compared with that of third day:t=49.74,P=0.02.After first day of treatment,the serum VEGF concentration was compared with that of one month:t=76.30,P=0.04.After third day of treatment,the serum VEGF concentration was compared with that of one month:t=35.09,P=0.00.Serum Arg-1 before treatment was compared with that after treatment: 1 day,t =-19.27,P = 0.01;3 day,t =-43.66,P = 0.04,1month t =-63.55,P = 0.00.After first day of treatment,the serum Arg-1 concentration was compared with that of third day:t=-35.93,P=0.04.After first day of treatment,the serum Arg-1 concentration was compared with that of one month:t=-67.98,P=0.04.After third day of treatment,the serum Arg-1 concentration was compared with that of one month:t=-22.74,P=0.01.Serum iNOS before treatment was compared with that after treatment: 1 day,t = 29.64,P = 0.04;3 day,t = 49.19,P = 0.03;1 month t = 64.71,P =0.01.After first day of treatment,the serum iNOS concentration was compared with that of third day:t=24.52,P=0.01.After first day of treatment,the serum iNOS concentration was compared with that of one month:t=43.64,P=0.03.After third day of treatment,the serum i NOS concentration was compared with that of one month:t=17.71,P=0.02.The serum i NOS and VEGF concentrations of patients with advanced NSCLC decreased continuously and the concentration of Arg-1 continued to rise.The difference was statistically significant(P<0.05).The positive rate of iNOS and VEGF r was 0.379 in peripheral blood of patients with NSCLC,and negative correlation between Arg-1 and VEGF r was-0.556,the difference was statistically significant.Conclusion:1.There was no significant difference in VEGF,Arg-1 and iNOS between peripheral blood serum and age,tumor histology in patients with advanced NSCLC.2.The concentrations of VEGF,Arg-1 and iNOS in peripheral blood of patients with NSCLC were significantly higher than those in healthy controls,the difference was statistically significant.3.There was a negative correlation between the expression of Arg-1 and VEGF in non-small cell lung cancer patients,and the expression of iNOS was positively correlated with VEGF expression.4.The levels of serum VEGF and iNOS 1 day,3 day,1 month after treatment in the NSCLC group were lower than those before treatment,and the serum Arg-1 concentration was higher than that before treatment,and the difference was statistically significant.Suggesting that microwave ablation is an effective treatment of cancer,it candirectly kill cancer cells,but also affect the vascular endothelial growth factor,arginase-1,inducible nitric oxide synthase levels,to a certain extent,changes in tumor microenvironment,induce antitumor immunity,thus affecting the growth of tumors,and provide experimental basis for clinical treatment and prognosis. |
