Objective:Preparation of BMP-2-loaded college-like protein hydrogel to repair bone defects in order to obtain good bone repair materials.Methods: Preparation of college-like protein(CL),oxidation of carboxymethyl cellulose(OCMC),N-succinyl chitosan(NSC),mixed to form the hydrogel.The physical,chemical properties and biological properties of microstructures and chemical structures were studied by SEM,FTIR and MTT experiments.44 rat skull defect model were prepared successfully,then grouped,the gel complex was implanted into the animal skull defect model to study the effect of the gel on bone repair.The rats were sacrificed at 2,4,8,12 weeks in time sequence,and the osteogenesis effect was studied by CT,MRI and histological staining.Results: 1.OCMC,NSC and CL were mixed to form gel and it has a good release performance.2.The animal model was prepared and the bone regeneration of the skull defect was detected by CT,MRI and histological sections at 2,4,8 and 12 weeks postoperatively.The results showed the bone tissue defect had repaired obviously.Conclusion: CL / OCMC / NSC compound BMP-2 gel is a new type of osteogenic material,with excellent character of bone regeneration.
