| The research of this paper is focused on classic Qing’e formula and compound Codonopsis.Qing’e formula(QEF)is a classic traditional Chinese medicine recipe comprised of Eucommniae Cortex,Psoraleae Fructus,Juglandis Semen and Garlic Rhizoma.The protection of each single herbs in Qing’e formula on learning and memory has been reported.However,the effects of QEF and its different compatibility on learning and memory have rarely been reported.Compound Codonopsis is comprised of five ingredients,Codonopsis pilosula,Angelica sinensis,Lycium barbarum,Radix Asparagi and wine Rheum officinale,Which has anti-aging and anti-oxidation ability.One purpose of this study is to observe the effects of different compatibility of QEF on the scopolamine-induced learning and memory impairment in the mouse,The other is to develop as a kind of antioxidant function of health care products for compound Codonopsis.Based on this,the major work of this paper may be summarized as the follow:(一)The effects of different compatibility of Qing’e formula on scopolamine-induced learning and memory impairment in the mouseThe extracts of QEF and its different compatibility were obtained as followed: a)EC;b)EC+ PF;c)EC + JS;d)EC + GR;e)EC + PF + JS + GR(QF);f)PF.Six different compatibility of QEF were infiltrated with 8-fold mass of ethanol-water(75:25,v/v),respectively,and then extracted,and administered with corresponding animals for 14 days.The results showed that different compatibility of QEF significantly prolonged latency in the step-down test(P < 0.05 or P< 0.01),shortened escape latency in the navigation test(P < 0.05 or P < 0.01),increased the residence time percent and swimming distance percent in the target quadrant in the probe trial session(P < 0.05 or P < 0.01).Additionally,our results also found that different compatibility of QEF remarkably inhibited AChE activity and increased BDNF expression in the hippocampus of mice.What’s more,the group after treated with whole recipe(QF)showed the highest level of improvement.These findings not only suggest that QEF may effectively ameliorate cognitive deficits via inhibiting AChE activity and increasing BDNF expression in the hippocampus,butalso elucidate the rationality of QEF.(二)Pharmaceutical research of compound Codonopsis capsule(1)Extraction technology research: orthogonal design method was employed for conducting the multiple guidelines evaluation and selecting the optimum water extraction technology with the contents of total polysaccharide,flavonoids and saponins and the yield of extraction as the detective index.The best water extraction technology was soaking for 4 h,adding 8,6,6 times and boiling 3 times with 1h for each time.(2)Extract scavenging free radicals research: the removal ability of Compound extract for free radicals was researched.The results found that the formula extracts on DPPH·,·OH,O2-· had a certain ability to remove free radicals,and the dose concentration is 0.04 ~ 1.23 mg/ml,present a dose dependent.(3)Dosage form selection and preparation process study: to extract and different materials consisting of a mixture of moisture absorption percentage of indicators to determine the type and proportion of auxiliary materials;For forming rate and granulating difficulty level indicators,screening preparation best molding process.As auxiliary materials,microcrystalline cellulose,dextrin was 1:3 and drugs: auxiliary ratio was 1.5:1,60% ethanol as moistening agent,the value of capsule of liquidity is lesser,meet the production requirements.Therefore optimizing the preparation process is: 1.5:1 to join accessories(microcrystalline cellulose: dextrin = 1:3),and60% ethanol as moistening agent,20 mesh granulating,80 ℃ drying,granulation,capsule.(4)The quality standard research: from qualitative identification,content determination and the general principles of the preparation check against oxidation care capsules for quality test research.Results showed that the thin layer chromatography(TLC)identification of Chinese angelica,barbary wolfberry fruit,rhubarb spots were clear,accurate,and negative no interference.All the inspection in line with the "Chinese pharmacopoeia" 2015 edition of four agents stipulated under the general principles of the capsule.Major component of reducing sugar content in preparation for: 23.20 g/100g;The flavonoids content for: 8.71 mg/100 g;The content of total saponins for: 1.20 g/100 g.(5)Antioxidant function evaluation: according to "health food inspection and evaluation of technical standards(2003 edition)" in antioxidant function test method,respectively,to study the compound codonopsis capsule on D-galactose to aging model mice and60Coγ irradiation malondialdehyde in serum and liver of mice MDA level,T-SOD and GSH-Px activity.Founding in D-galactose senile model,compound of Dangshen capsule in middle dose(1.2 g/kg),low dose(0.6 g/kg)could obviously increase the T-SOD activity in serum and liver of mice(P < 0.05 or P< 0.01),and significantly improve the activity of GSH-Px in serum(P < 0.05 or P < 0.01),significantly reduce the MDA content in liver tissue(P < 0.01);High dose(2.4 g/kg)can significantly reduce the MDA content in serum and liver(P < 0.05 or P < 0.01),and at the same time improve T-SOD activity in serum(P < 0.01).In60Coγ radiation model,compound codonopsis capsule high dose can significantly increase GSH-Px activity in serum and liver of mice(P < 0.05 or P < 0.01),and at the same time also can significantly reduce the MDA content in mice liver model(P < 0.01)and increase T-SOD activity(P < 0.05).Middle dose can significantly improve the liver GSH-Px activity and reduce MDA content(P < 0.01).According to health food antioxidant function evaluation criteria to judge the animal experimental results "any indicators in the lipid peroxide content and antioxidant enzyme activity in any indexes is positive,can determine the antioxidant animal experiment result positive subjects sample",confirmed that the compound codonopsis capsule with antioxidant health care function. |
