Objectives To analyze the effect of pentamethyl quercetin(PMQ)on the expression of Nrf2 and HO-1 in hippocampus of diabetic rats.Methods 1 SPF grade 13 weeks male GK rats 60,were randomly divided into 5 groups:MG group(GK model group,n = 12),HL group(PMQ low dose group,n = 12),HM group(PMQ middle dose group,n = 12),HH group(PMQ high dose group,N = 12),LG group(lipoic acid group,n = 12).We selected homologous healthy Wister rats as NG group(normal group,n = 12).HL group: given low dose of PMQ(PMQ 2.5mg / kg body weight);HM group: given medium dose of PMQ(PMQ 5mg / kg body weight);HH group:given high dose of PMQ(PMQ 10 mg / kg body weight);LG group given lipoic acid(lipoic acid 60 mg / kg body weight);NG,MG group: given equal volume of dimethyl sulfoxide;2 The body weight、FBG was measured before administration,administration for 8 weeks.The histological changes of the hippocampus were detected by HE staining.The expression of Nrf2 and HO-1 in the hippocampus of rats was detected by immunohistoch-emistry and immunoblotting.Results 1 Weight test results: Weight before treatment: There was no significant difference in body weight between the each group of rats(P>0.05).8 weeks after treatment Weight: Compared with NG group rats,MG group had lower body weight,the difference was statistically significant(P<0.05);The body weight of rats treated with PMQ increased gradually with the increase of drug concentration.The effect of HH group was the most significant(P<0.05).Compared with LG group,the body weight of HH group was not statistically significant(P>0.05).2 FBG test display: FBG results before administration:Compared with NG group,the FBG of the other groups was higher than that of NG group,the difference was statistically significant(P<0.05),but there was no significant difference between the other groups(P>0.05).8 weeks of administration of FBG results: Compared with NG group,FBG was significantly higher in MG group(P <0.05).The FBG of rats treated with PMQ decreased gradually with the increase of drug concentration,The effect of HH group was the most significant,the difference was statistically significant(P<0.05);There was no significant difference in FBG between the HH group and the LG group(P>0.05).3 HE staining showed: The neurons of CA1 in hippocampus of NG group were all arranged.The neuronal cells in the hippocampus of the MG group were disordered,the cell membrane was wrinkled,Deletion and cytoplasm reduction.PMQ intervention in rat hippocampus CA1 region after the arrangement of nerve cells than the pre-structured,rich before the cytoplasm,HH group of nerve cells,the most complete capsule,the most abundant cytoplasm,LG group and HH group compared to the difference was not obvious.4 Immunohistochemical test results: The positive expression of Nrf2 protein in the CA1 region of the hippocampus was cytoplasm or nucleus,and the Nrf2 protein in the NG group was expressed in the cytoplasm,and no significant expression was observed in the nucleus(P<0.05).Compared with NG group,the expression of Nrf2 protein in MG group was increased(P<0.05).After treatment with PMQ,the expression of Nrf2 protein was further enhanced(P<0.05).Compared with HH group,There was no significant difference in expression(P>0.05).The positive expression of HO-1 protein in rat hippocampal CA1 region showed brown granules in cytoplasm.The expression of HO-1 protein in NG group was small in cytoplasm.Compared with NG group,HO-1 protein in MG group increased in cytoplasm(P<0.05).After the PMQ intervention,the expression of HO-1 proteinincreased gradually with the increase of drug concentration(P<0.05).Compared with HH group,there was no significant difference in the expression of HO-1 protein in LG group(P>0.05).5 Immunoblot test results: Nrf2 total protein expression: NG group<MG group<HL group<HM group<HH group(P<0.05),HH group=LG group(P>0.05).Nrf2 nucleoprotein expression: NG group<MG group<HL group<HM group<HH group(P<0.05),HH group=LG group(P>0.05).HO-1 protein expression: NG group<MG group<HL group<HM group<HH group(P<0.05),HH group=LG group(P>0.05).Conclusions 1 PMQ has an improved effect on metabolic disorders such as decreased weight and hyperglycemia in diabetic rats.2 PMQ can improve the damage of nerve cells in hippocampus of diabetic rats.3 Quercetin can enhance the expression of Nr F2 and HO-1 proteins in the hippocampal CA1 neurons of diabetic rats.
