This dissertation consists of four chapters.The first chapter elaborats the investigation on the chemical constituents of the root of Swertia kingii Hook.F.The second chapter reports the analysis of main components in Swertia kingii Hook.F,Gentiana tibetica King ex Hook.f.and Gentiana dendrologi H.Smith by HPLC-ESIMS.The third chapter reports the optimization of extraction technology of total xanthone from Swertia kingii Hook.F.The fourth chapter mainly reviews the Swertia research progress on chemical constituents and pharmacological activities of plants.The first chapter reports the isolation of twenty-one chemical constituents from the 95% ethyl acetate extract of Swertia kingii Hook.F root by silica column chromatography,macroporous resin,Si60 column chromatography,C18 column and Sephadex LH-20 and recrystallization.21 compounds were isolated from Swertia kingii Hook.F.By determination of physicochemical constants,combined with IR,UV,ESI-MS,1H-NMR and 13C-NMR,the compounds were identified as swertianolin(1),1,3,7,8-tetrahydroxyxanthone-1-O-?-D-glucopyranoside(2),demethylbellidifolin(3),1,3,7,8-tetrahydroxy-xanthone(4),1,2,6,8-tetrahydroxy-xanthone(5),1,3,7-trihydro-xanthone(6),gentiacaulein(7),1,3,7-trihydrox-8-methoxyxanthone(8),1,5,8-trihy drox-3-methoxyxanthone(9),1-hydrox-3,7,8-trimethoxy-xanthone(10),isoorientin(11),methylswertianin(12),mangiferin(13),diisobutylphthalate(14),sweroside(15),gentiopicroside(16),swertiamarine(17),oleanolic acid(18),daucosterol(19),?-sitosterol(20),sucrose(21).The 21 compounds included xanthone and its glycosides,flavonoid C-glycosides,secoiridoid glycosides,aromatic acid,sterols,triterpenoids and disaccharide.Compounds 1~6,8~10,14~19 and 21 were isolated from the root of Swertia kingii Hook.F for the first time.The second chapter consists of two parts.The focus of the first part was the determination of the main components in the root of Swertia kingii Hook.F,the flowers of Gentiana tibetica King ex Hook.f.and Gentiana dendrologi H.Smith by HPLC-ESI-MS.The focus of the second part was the determination of the eight components in the root of Swertia kingii Hook.F by HPLC.The third chapter includes of two parts.The first reports the determination of total xanthones from the root of Swertia kingii Hook.F.Using the Al(NO3)3 method,the measured total xanthones content was 13.24%.The focus of the second part was optimization of the extraction process of xanthones from the root of Swertia kingii Hook.F.by single-factor and response surface methodology.The best solution is: the concentration of sample solution was 1mg/m L,the flow rate was 1 m L/min,p H 5 under the condition of the sample,the adsorption rate 84.16%;in 50% of eluent concentration,eluent flow rate 3.5m L/min,elution experiment liquid was 11 BV conditions of elution,the elution rate was 48.22%,the measured total xanthones content was 41.57%,the refining rate can reach 314.1%.The fourth chapter mainly introduces the method of the Swertia plants chemical composition,pharmacological activity and effective components extraction,separation and purification,on the domestic and foreign research status of the Swertia plants were reviewed searched by Sci Finder ? CAPLUS and MEDLINE et al.databases.
