Objective: To detect the expression level of plasmacytoma variant translocation 1(PVT1),a long non-coding RNA in blood and tissues of gastric cancer(GC)patients and normal persons.Analyze the diagnosis value of PVT1 in GC and discuss the effects of PVT1 and c-myc on the development of GC.Methods:(1)Collected 87 blood samples from GC patients and 94 blood samples from normal control.Used quantitative real-time PCR(q RT-PCR)technology to detect blood m RNA level of PVT1 in blood.(2)Collected 50 GC patients and 40 normal gastric mucosa of paraffin specimens.The expression of PVT1 and c-myc protein in gastric cancer were detected by in situ hybridization and immunohistochemistry.χ2 test were used to analyze the expression level of PVT1 and c-myc in tissues and the correlation with the clinical characteristics.Spearman test was used to analyze the correlation between PVT1 and c-myc.(3)Silenced the expression of PVT1 through RNA interference(RNAi)technology,and detected the effect of PVT1 on the proliferation,invasion and migration of GC cells using CCK8,transwell invasion and migration experiments.(4)Western Blot was used to detect the relationship between PVT1 and c-myc.Results:(1)The expression of PVT1 in GC blood was higher than that of the normal group which was correlated with lymph node metastasis and clinical stage(P<0.05).(2)ISH and IHC showed that PVT1 and c-myc were expressed both in cytoplasmic and nuclei,and were highly expressed in GC tissues(P<0.05).The expression level of PVT1 was correlated with lymph node metastasis and tumor stage(P<0.05).The expression of c-myc protein was correlated with clinical stage of GC patients(P<0.05).Spearman correlation analysis showed that PVT1 was positively correlated with c-myc(P<0.05).(3)Silienced the expression of PVT1,the invasion and migration ability of BGC823 and AGS GC cells were decreased,but the proliferation ability was not changed.(4)Western Blot results showed that the expression of c-myc protein was decreased when silenced the expression of PVT1.Conclusion: PVT1 is highly expressed in the blood and tissues of GC patients and is associated with lymph node metastasis and clinical staging.Inhibition of PVT1 expression in gastric cancer cells,the invasion and migration ability is decreased,PVT1 expression and c-myc expression were positively correlated,PVT1 may be affected the expression of c-myc participate in the development of gastric cancer.
