Objective To observe the expression and function of long non-coding RNA myocardial infarction associated transcript(LncRNA-MIAT)in TNF-α stimulated vascular inflammation in vitro.Methods The expression of intercellular adhesion molecule-1(ICAM-1),vascular cell adhesion molecule-1(VCAM-1)and LncRNA-MIAT was detected respectively by Western Blot,PCR and qRT-PCR in ECs and VSMC under the inflammatory station induced by TNF-α.Moreover,the expression of ICAM-1 and VCAM-1 sitimulated by TNF-α was detected in the ECs and VSMC transfected with siRNA MAIT.The migration of ECs and VSMC transfected with siRNA MAIT was observed after sitimulated by TNF-α.Results 1.(1)PCR result of endothelial cell:Contrast with no primer,add primer of lncRNA-MIAT show high brightness.No difference be found between different concentrations of lncRNA-MIAT primer.Indicate that endothelial cell experess lncRNA-MIAT.(2)PCR result of vascular smooth muscle cell:Contrast with no primer,add primer of lncRNA-MIAT show high brightness.No difference be found between different concentrations of lncRNA-MIAT primer.Indicate that vascular smooth muscle cell experess lncRNA-MIAT.2.(1)qRT-PCR result of endothelial cell : LncRNA-MIAT expression level was significantly up-regulated under the stimulation of TNF-α,and with a time-and dose-dependent pattern in endothelial cell.(2)qRT-PCR result of vascular smooth muscle cell:LncRNA-MIAT expression level was significantly up-regulated under the stimulation of TNF-α,and with a time-and dose-dependent pattern in vascular smooth muscle cell.3.(1)RNA interferrence result of endothelial cell:LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the expression of LncRNA-MIAT compared with the control after 48 h stimulated by TNF-α.(2)RNA interferrence result of vascular smooth muscle cell:LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the expression of LncRNA-MIAT compared with the control after 24 h stimulated by TNF-α.4.(1)Western Blot result of ICAM-1 in endothelial cell:LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the expression of ICAM-1 compared with the control after 24 h stimulated by TNF-α.(2)Western Blot result of VCAM-1 in endothelial cell:LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the expression of VCAM-1 compared with the control after 24 h stimulated by TNF-α.(3)Western Blot result of ICAM-1 in vascular smooth muscle cell: LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the expression of ICAM-1 compared with the control after 24 h stimulated by TNF-α.5.(1)Migration test result of endothelial cell : LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the migration of endothelial cell after 24 h stimulated by TNF-α.(2)Migration test result of vascular smooth muscle cell:LncRNA-MIAT knockdown by siRNA MAIT significantly decreased the migration of vascular smooth muscle cell after 24 h stimulated by TNF-α.Conclusion LncRNA-MIAT may be involved in the vascular inflammatory,and serve as a pro-inflammatory factor.
