Objective: To detect the expressions of lnc RNA-PRSS3P2,PRSS3-m RNA and PRSS3 protein in papillary thyroid carcinoma(PTC)and to explore the clinicopathologic significance of PRSS3 and PRSS3P2 in PTC.To research the impact of interfering PRSS3P2 on the biological function of TPC-1 cells.Methods: 1)The expressions of PRSS3-m RNA and lnc RNA-PRSS3P2 were detected by real-time quantitative PCR in 55 PTC fresh tissues.The relationship between expressions of PRSS3/PRSS3P2 and the clinicopathologic features were carefully analyzed.2)TPC-1cell line was infected by the PRSS3P2-sh RNA cloned lentivirus vector,and the expression of lnc RNA-PRSS3P2 was detected by real-time quantitative PCR.And cell proliferation of TPC-1cell was analyzed by CCK-8 assay.3)The expression of PRSS3 protein was measured by immunohistochemical assay in 55 PTC tissues plus with their paired normal thyroid tissue.The relationship between the expression of PRSS3 protein and clinicopathological characteristics was analyzed.Results: 1)The expression levels of PRSS3 and PRSS3P2 in PTC tissues were obviously higher than their non-tumor counterparts(P<0.05).PRSS3P2 was correlated with the age of patients(P<0.05);However,PRSS3 is not associated with the clinicopathologic features of PTC.2)The expression level of PRSS3P2 in transfected TPC-1 cells was lower than that of untransfected cells(P<0.05),but no significant change was found in cell proliferation ability(p>0.05).3)The expression level of PRSS3 protein in PTC tissues was higher than paired non-tumor counterparts and the expression of PRSS3 protein was correlated with lymph node metastatisis(P<0.05).Conclusion: The expressions of PRSS3-m RNA and lnc RNA-PRSS3P2 were increased in PTC.The present findings indicate that PRSS3 and PRSS3P2 may participate the development of PTC.PRSS3-m RNA and lnc RNA-PRSS3P2 can be served as two potential diagnostic markers for PTC.
