Objective: To investigate the expression of NCAM and β-catenin in thyroid cancer cells,and the difference of β-catenin and NCAM expression between the thyroid carcinoma tissue and the benign thyroid lesions,so as to investigate the significance of NCAM andβ-catenin in development of thyroid cancer,as well as their significance in the metastasis,the invasion of thyroid cancer.Methods: Paramn—embedded tissue blocks of 70 cases of thyroid carcinoma,36 cases of benign thyroid tumor from Jau.2008 to Oct.2015,were obtained from Linyi People’s Hospital.All cases were diagnosed clearly,the clinical datas were complete and no special treatment was given before operation.To investigate the relationship between expression of NCAM and β-catenin and the clinical pathological datas,NCAM and β-catenin molecules were detected by immunofluorescence staining method.The immunological reagents were employed monoclonal antibody against of NCAM and β-catenin.All statistical analyses were done using SPSS software,version 19.0 by X 2 test,Spearman rank correlation analysis.Differences were considered signifcant at P-values <0.05.Results:1.In the tissue of thyroid benign lesions,β-catenin protein mainly expressed in the cell membrane,while β-catenin protein expressed in the cytoplasm or nucleus of ectopic tissue in thyroid cancer tissues,the positive expression in the cell membrane was significantly lower than that in benign thyroid tissue.The positive expression ofβ-catenin in thyroid carcinoma group were significantly higher than that in the benign thyroid lesions group(all P<0.05).The abnormal expression rates of β-catenin in different histological types of thyroid carcinoma,including papillary thyroid carcinoma,thyroid follicular carcinoma,medullary thyroid carcinoma and anaplastic thyroid carcinoma,was not statically significant(all P>0.05).The aberrant expression of β-catenin in the thyroid carcinoma tissue was not associated with sex、age、diameter of tumor、lymph node metastasis and TNM stage(all P>0.05).2.The positive expression of NCAM was mainly located in the cell membrane.The positive expression rate of NCAM in thyroid carcinoma was lower than that in benign thyroid lesions(P<0.05).The difference of positive expression rate of NCAM in different histological types of thyroid carcinoma tissues has no statistical significance(P>0.05);the positive rate of NCAM in lymph node metastasis,were statistically significant(P<0.05).The decreased expression of NCAM and the aberrant expression of β-catenin in the thyroid carcinoma tissue was not associated with sex、age、diameter of tumor and TNM stage(all P>0.05).3.The positive expression of NCAM and the abnormal expression of β-catenin in thyroid carcinoma were negatively correlated with Spearman(r=-0.025,P<0.05).Conclusions: 1.β-catenin in thyroid cancer are common ectopic expression,the expression of cell membrane decreased and cytoplasm or nucleus expression increased,suggesting that the abnormal expression of β-catenin may play an important role in the occurrence and development of thyroid carcinoma;2.The positive rate of NCAM in thyroid cancer compared with benign thyroid lesions decreased significantly;the expression rate of NCAM was closely related with lymph node metastasis,suggesting that NCAM may be play an important role in thyroid carcinoma molecular signal pathway in metastasis and invasion of thyroid carcinoma;3.Combined detection of β-catenin and NCAM may help for differentiating thyroid carcinoma with benign thyroid lesions;while there was little value in the diagnosis of undifferentiated thyroid carcinoma and papillary thyroid carcinoma,thyroid follicular carcinoma,medullary thyroid carcinoma.4.The expression of β-catenin and NCAM protein was negatively correlated and the reverse balance suggest that NCAM and β-catenin are closely related to the occurrence,development,invasion and metastasis of thyroid carcinoma...
