Chinese medicine Yizhiren, also known as Yizhizi, is the fruit of ginger family,perennial herb (Alpinia oxyphylla Miq.), is one of the traditional Chinese medicine,included in the "Chinese Pharmacopoeia" 2015 edition . It is the first batch 101 races of medicine and food homologous. It is one of four south medicine of China. It can supply kidney, take saliva, solid fine, shrink urine. Supplying kidney drugs have more neurological protection, improve the role of intelligence. There is a record for improving memory and it is called as "Yizhi". Therefore, to reduce neurological impairment after cerebral ischemia and promote the recovery of neurological function is the focus of treatment of cerebral ischemia. Therefore,we selected the active ingredients in the puzzle separation and identification and neuroprotective efficacy screening, and to explore the mechanism.Through the early literature research, from the benefits of the selection of the chrysin, nootkatone, tectochrysin three kinds of major, potential ingredients. The results of the screening showed that the optimal concentration of chrysin was 1μM,and the NF promoter could be up-regulated by 2.21 times. The optimum concentration of nootkatone is 1μM, which increases the NF promoter by 2.3 times. The optimal concentration of tectochrysin is 1 μM, which increases the NF promoter by 1.6 times.The bone marrow mesenchymal stem cells were treated with different concentration gradient and time gradient. The results of real-time fluorescence quantitative PCR showed that the two kinds of monomers could promote the transcription of five kinds of nerve markers to a certain extent at a concentration of 1μM and reflect a certain dose-dependent. The transcription level of mRNA reached a peak at 72 h, and the effect was superior to that of the selected positive control (all-trans retinoic acid).Immunofluorescence experiments showed that the proteins corresponding to the five neurotransmitters were also detected when bone marrow mesenchymal stem cells were induced for 72 h. Therefore, it can be concluded that these two compounds have a certain ability to induce bone marrow mesenchymal stem cells to differentiate into neuronal cells.This study found two compounds that could induce stem cell differentiation into neurons from Chinese medicine, and found the material basis for the neuroprotection of the puzzle and the ability to promote nerve regeneration. It provided an example for the rapid screening of small molecule compounds from traditional Chinese medicine , For the modernization of traditional Chinese medicine and Yizhi Ren quality standards to provide a reference.
