Objective: Observation and analysis of Quyujiedu of endometriosis(endometriosis,EMS)key effector molecules in Nrf2/ARE pathway in patients with Nrf2(NF-E2-related factor 2),superoxide dismutase(superoxide dismutase,SOD)expression of intervention,starting from the perspective of oxidative stress,looking for possible targets Quyu Jiedu formula played against oxidative stress,try to make some explanation on the antioxidant mechanism.Methods: Methods 66 cases of EMS patients met the study criteria in this experiment,were randomly divided into experimental group(Quyujiedu granule +IVF)33 cases and observation group(Guizhi Fuling Capsule +IVF)33 cases.At the same time,33 patients(+IVF)were selected as the control group because of the factor of IVF.Three groups of patients were treated with each group of drugs for three months.Comparison between the experimental group and the observation group before and after treatment,blood stasis and toxin accumulation syndrome score and serum carbohydrate antigen 125(CA125)levels were compared between the three groups of oocytes,fertilization rate,embryo number,rate of high quality embryos,using quantitative real-time fluorescence(RT-PCR)expression of Nrf2 and SODmRNA in granulosa cell technology;Western blot(Western-blot)method for the determination of the expression of Nrf2 and SOD protein in granulosa cells;thiobarbituric acid method(TBA method)determination of MDA in granulosa cells.Result: 1.The experimental group and the observation group after treatment,blood stasis and toxin accumulation syndrome integral were decreased,with statistical significance(P < 0.05),the experimental group decreased much more than those in the observation group,was statistically significant(P < 0.05)2.Three groups of oocytes,good quality embryos,fertilization rate,good embryo rate comparison: experimental group than the control group,the observation group of oocytes,good quality embryos,fertilization rate,good embryo rate,were statistically significant(P < 0.05);the experimental group compared with the observation group number of embryo and fertilization rate good embryo rate is high,the more close to the control group,the difference was statistically significant(P < 0.05);the experimental group than in the observation group the number of oocytes is slightly higher,more close to the control group,but the difference was not statistically significant(P > 0.05)3.Three groups of patients with ovarian granulosa cells in the expression of Nrf2 and SOD were statistically significant(P < 0.05): the control group than in the experimental group,the observation group was down regulated in the experimental group than in the observation group4.Three groups of patients in the MDA content of granulosa cells were statistically significant(P < 0.05): the control group was lower than the experimental group and the observation group,the experimental group was lower than the observation group,more close to the control group.Conclusion: 1.Quyujiedu method can significantly improve the IVF cycle of EMS patients with blood stasis and toxin accumulation syndrome,reduce serum CA125 content,increased with fertilization rate,number of embryo and embryo rate 2.Patients with endometriosis had higher oxidative stress than normal subjects 3.Quyujiedu may through the Nrf2/ARE pathway,with Nrf2 as the target,improve the expression of key effector molecules in ovarian granulosa cells Nrf2,SOD gene and its related protein,reduced the content of MDA in granulosa cells,thus inhibiting the oxidative stress of egg cells,remove toxic harmful substances to the egg cell,thereby improving the egg cell the quality and the quality of embryo4.Quyujiedu may inhibit oxidative stress via the Nrf2/ARE pathway,part of the interpretation of the "Quyujiedu Tongluozhitong treatment," the science and rationality,enrich the study of blood stasis and toxin accumulation theory. |
