| Objective (s):The incidence and mortality of lung cancer top the list worldwide.However, the molecular mechanism of lung cancer has yet been elucidated.ZBED1 emerges as a crucial transcriptional factor correlated with cell proliferation. Currently, no research about ZBED1 involving in lung cancer has been reported. The aim of this study is to analyse the expression and effect of ZBED1 in lung adenocarcinoma cells and tissues,so as to provide the experimental evidence for the analysis of pathogenesis of lung adenocarcinoma.Methods:We used HE and IHC to detect the expression of ZBED1 in lung adenocarcinoma tissues and corresponding normal lung tissues and further analysed the correlation between ZBED1 and clinicopathological parameters.Westernblotting was used to analyse the expression of ZBED1 in lung adenocarcinoma cell lines as A549, PC-9, NCI-H1975, NCI-H2342,SPC-A-1, XLA-07, and normal lung epithelial cell as BEAS-2B.siRNA technique was used to knock down ZBED1 in lung adenocarcinoma A549 cell line.The effect of ZBED1 in lung adenocarcinoma cell proliferation was analysed using MTS assay and flow cytometry.The change of cell cycle related proteins were detected by Western blotting after the ZBED1 knock-down.Flow cytometry was used to analyse the relationship between ZBED1 and adenocarcinoma cell apoptosis.Results:The expression of ZBED1 in lung adenocarcinoma tissues was higher than that in corresponding normal lung tissues.ZBED1 was expressed in many lung adenocarcinoma cell lines, with A549 cell line the highest.The proliferative ability was decreased after ZBED1 was knocked down in lung adenocarcinoma cell line A549.After ZB EDI was knocked down, there was a G0/G1 phase arrest in A549 cell line, and Westernblotting results showed that the expression of CyclinD3, CDK4, and CyclinEl was decreased,P15, P27, and P53 was increased,while there was no significant change of the expression of CyclinD1,CyclinD2, CyclinE2, CDK2, CDK6, and P21.The apoptosis rate had no statistically significant change in A549 cell line after ZBED1 was knocked down.Conclusion(s):ZBED1 was highly expressed in lung adenocarcinoma. ZBED1 knock-down may inhibit the lung adenocarcinoma cell proliferation through cell cycle arrest. |
