| Dendranthema morifolium is the dry capitulum of compositae Chrysanthemum(Dendranthema morifolium(Ramat.)Tzvel.).The Han Dynasty "Shen Nong’s herbal classic" records: " D.Morifolium is bitter,flat in taste.which treates wind vertigo,gall,eyes wants to be taken off,tears out,the skin dead hunger and rheumatism rheumatism.It is benefit for blood and can prolong life for a long time to eat it." D.Morifolium mainly contains the effective components of flavonoids,terpenes,volatile oil,organic acids and polysaccharides,vitamin C,mineral elements,amino acids and other nutrients.Which exhibits antioxidation,antitumor,anti-inflammatory,antibacterial,hepatoprotective,antiviral,reducing blood lipid,enhance immunity and other pharmacological effects.2500 years ago,D.morifolium had been cultivativated in China,as recorded in "Liji·Yueling" "Chuci·Lisao"."Jupu of Kaifeng" had recorded 1606 species of D.morifolium,according to "Dongjingmenghualu".Which was not only for cultivation and watching,but also for eating in Kaifeng.However,there were few reports on the antioxidant activity and chemical composition of D.morifolium cultivated in Kaifeng.In order to elucidate the antioxidant efficacy material basis,studies on the antioxidant constituents of Dendranthema morifolium.cv.‘jinsidaju’were established using Bioassay-guided Method.In our research,15 kinds of compounds had been isolated from D.Morifolium.cv.‘jinsidaju’ by silica gel,TLC,ODS and HPLC.Their structures were identified by IR,UV,ESI-MS,HR-ESI-MS,1D and 2D NMR as follows:(6R,7R,9E)-7-hydroxy-11-methoxy-bisabolane-2,9-diene-4-one(jinsidajuol A,1),(6R,7R,9E)-7-hydroxy-bisabolane-2,9,11-triene-4-one(jinsidajuol B,2),(3R,7R,9R)-3,9-dihydroxygermacra-4(15),10(14),11(12)-triene(3),cyperusol C(4),β-sitosterol(5),naringenin-7-O-β-D-glucopyranoside(6),luteolin-7-O-β-D-(6″-acetyl)-glucoside(7),(2R)-eriodictyol-7-O-β-D-glucopyranoside(8),apigenin-7-O-β-D-glucoside(9),apigenin-7-O-β-D-(6"-O-acetyl)-glucoside(10),luteolin-7-O-β-D-glucoside(11),huazhongilexone-7-O-β-D-glucopyranoside(12),apigenin(13),chrysosplenol D(14),kaempferol-3-O-α-L-rhamnoside(15).Compounds(1-15)were all first isolated from the Chrysanthemum cultivars(D.morifolium.cv.’jinsidaju’)cultivated in Kaifeng.Compound 1 and 2 were two new compounds,14 containing polymethoxy flavonoid aglycones,had been reported in the Viticis fructus [118],D.Artemisia annua L.[119],D.Rudbeckia hirta L.[120],D.Centipeda minima [121] and other genera,but no genus have been reported in the Morifolium,so compound 14 is isolated from D.morifolium for the first time.The vitro antioxidant activity was evaluated by scavenging DPPH and ABTS free radicals.IC50 of 7,8,11,12,14 and BHT were 0.402 ± 0.079,0.515 ± 0.015,2.255 ± 0.279,0.528 ± 0.073,0.428 ± 0.066,0.680 ± 0.121,respectively in DPPH method,antioxidant activity was 7 > 14 > 8 > 12 > BHT >11;IC50 of 6,7,8,11,12,13,14,15 and BHT were 0.069 ± 0.037,0.117 ± 0.009,0.036 ± 0.006,0.375 ± 0.005,0.042 ± 0.004,0.111 ± 0.016,0.690 ± 0.333,0.757 ± 0.025,0.083 ± 0.004,respectively in ABTS method,antioxidant activity was 8 > 12 > 6 > BHT >13 > 7 > 11 > 14 > 15.Compound 8 had stronger ability than BHT in scavenging free radical activities in both method.In order to clarify the pharmacological basis of its antioxidant activity,promote the development and utilization of Kaifeng D.morifolium and provide a theoretical basis and scientific guidance for the development of the local economy,the separation of chemical constituents of D.morifolium and antioxidant activity in vitro were conducted by bioassay-guided method,... |
