Background:The side effects caused by the use of disinfectants,especially chemical disinfectant external environment pollution has raised the great attention.Researchers have long been searching for and research a kind of disinfectant with the advantages of good sterilizing effect,no toxic side effect and no pollution to environment.The main research directions are technology of disinfectant compound,synergistic effect of action factors,and a variety of new preparation technology,etc.,With the rapid development of biotechnology,biological products,such as Chinese herbal medicine plant extracts,enzymes,antibacterial peptide,phage and so on,began to be researched in the field of disinfection.Because these biological products,compared with the traditional disinfectant,have less irritation,pollution,adverse effects,they gradually are developed into new biological di sinfectant.Objective:We chose a dissolve staphylococcus enzyme and antibacterial peptide as the main ingredients of compound disinfectant.Our purposes are to observe the bactericidal effect of disinfectant,stability,poison rational and skin disinfection effect,and provide the scientific evidence for the practical disinfection of the complex biological disinfectant.Methods:Suspension quantitative germicidal test(including the neutralizer appraisal test)was performed to study the complex biological disinfectant effect of killing microorganisms,and validate the practical dose of disinfectant and sterilization ability.We used the test strains of staphylococcus aureus ATCC 6538(bacteria breeding of streptococcus pyogenes representative),pseudomonas aeruginosa ATCC 15442(the most common bacteria in nosocomial infection of propagule representative),candida albicans ATCC 10231(on behalf of pathogenic fungi).For toxicology test,we chose experimental animal and observed toxic effects of the disinfectant on experimental an imals.Using disinfectant rival disinfection field test,the natural bacteria killing effect on the surface of the disinfectant rivals,to verify that the disinfectant disinfection practical dose.A field experiment was conducted to study the germicidal efficacy of the disinfectant against the natural bacteria on the surface of skin.Results:The compound biological disinfectant(whose main ingredients are dissolve staphylococcus enzyme 20 u/ml,antibacterial peptide 0.5 mg/ml)can kill staphylococcus aureus,pseudomonas aeruginosa and candida albicans in the suspension after it was applied to suspension within 5 min,its killing log values were greater than 5.00,thus it can effectively kill the common pathogenic bacteria;Having been sprayed onto 30 persons’ skin for 5 min,the disinfectant liquid can kill the natural bacteria,its killing log values were greater than 1.00,based on which we can conclude that it can kill natural bacteria on the surface of human skin efficiently;The disinfectant can be stored on a constant temperature condition of 37 ℃ for 90 days,and when used after storage,it can still kill candida albicans within 5 min,its killing log value was greater than 4.00,consequently it will be valid for 2 years;In the mice acute oral toxicity test where this disinfectant liquid was used,LD50 were greater than 5000 mg/kg.bw,proving it is non-toxic,for rabbits,skin irritation index is 0,so that its stimulation intensity level can be sorted as non-irritating,and for guinea pigs,skin sensitization rate was 0%,therefore we can safely draw a conclusion that there isn’t any skin allergy reaction.Conclusion:The compound disinfectant has good sterilization effect on a variety of pathogenic bacteria,and have long-term stability as well,which belongs to a kind of non-toxic material.There is no sensitization effect for skin,and is an effective,safe,and ideal biological disinfectant.As a consequence,it should be further developed and we ought to expand its range of application,realizing its utility valve more extensively.
