| This thesis consists of three chapters.The first chapter determinated change rules of six secondary metabolites(salidroside,ligustroflavone,specnuezhenide,oleuropein,oleanolic acid and ursolic acid)in Ligustri Lucidi Fructus and Ligustri Lucidi leaves by reversed phase high-performance liquid chromatography,and it provided a theoretical basis for efficient use of Ligustri Lucidi Fructus and Ligustri Lucidi leaves.The second chapter investigated three anti-oxidation methods in eliminating1,1-Diphenyl-2-picrylhydrazyl radical,2,2’-Azinobis-(3-ethylbenzthiazoline-6-sulphonate)and Fe3+to analyze the antioxidation capacity of Ligustri Lucidi Fructus and Ligustri Lucidi leaves,we found the relationship between the secondary metabolites and antioxidant capacity in growth periods by partial least squares was established.The third chapter investigated four kinds of heavy metal including lead(Pb),cadmium(Cd),mercury(Hg)and arsenic(As)contents In different periods of Ligustri Lucidi Fructus and Ligustri Lucidi leaves by Inductively Coupled Plasma Optical Emission Spectrometer,and provided guidance for the safety of medicine.Chapter 1 Studies on Temporally Dynamic Change of Secondary Metabolites in Ligustrum lucidum Ait During Fruit and Leaf GrowthThis chapter established a method for studying on temporally dynamic change of salidroside, ligustroflavone,specnuezhenide,oleuropein,oleanolic acid and ursolic acid in Ligustrum lucidum Ait during fruit and leaf growth.Using methanol as the extraction solvent and ultrasound-assisted extraction combined with RP-HPLC.Single-factor analysis was adopted to find influence of different extraction solvents,mesh number,Solid-to-liquid ratios,ultrasound times and centrifugation speeds.The content of secondary metabolites was different in ligustrum lucidum during fruit and leaf growth,on the whole,Ligustri Lucidi Fructus display a downward trend and Ligustri Lucidi leaves display a rising trend.Chapter 2 Spectrum-effect relationship in antioxidant activity of Ligustrum lucidum Ait During Fruit and Leaf GrowthThis chapter explored the relationship of the active ingredients and antioxidant activity of Ligustrum Lucidum Ait during fruit and leaf growth by HPLC Three anti-oxidation methods,including DPPH,ABTS and FRAP,were used to analyze the antioxidation capacity.Ligustri Lucidi Fructus of antioxidant activity was also decreased with time passing,but Ligustri Lucidi leaves of antioxidant activity was also increased with time passing.There was a relationship between the content of secondary metabolites and the antioxidant activity.Using multipoint algorithm based on least-mean-square method about the peak,28 peaks were matched for Ligustri Lucidi Fructus,S2,S4,S8,S11,S12,S13,S14,S15,S16,S17,S19,S23,S25,S27,and S28 were positively correlated to opposite number of IC50 value in DPPH.S1,S2,S4,S5,S6,S9,S10,S11,S12,S13,S14,S16,S17,S19,S22,S23,S25,S27 and S28 were positively correlated to opposite number of IC50 value in ABTS.S1,S2,S11,S12,S13,S14,S15,S17,S19,S20,S22,S23,S25 and S28 were positively correlated to equivalent Trolox in FRAP.Salidroside,ligustroflavone,specnuezhenide,oleuropein,oleanolic acid and ursolic acid correspond to S11,S22,S23,S25,S27 and S28,respectively.42 peaks were matched for Ligustri Lucidi Leaves,S2,S4,S5,S6,S7,S8,S10,S12,S13,S14,S15,S16,S17,S18,S19,S20,S21,S22,S23,S24,S25,S26,S27,S28,S29,S30,S32,S38 and S40 were positively correlated to opposite number of IC50 value in DPPH,S2,S4,S10,S12,S13,S14,S15,S17,S19,S20,S21,S22,S24,S25,S27,S28,S29,S30,S33,S38 and S40 were positively correlated to opposite number of IC50 value in ABTS.S3,S5,S6,S8,S9,S10,S12,S13,S14,S15,S17,S18,S20,S21,S26,S28,S29,S30,S31,S32 and S38 were positively correlated to equivalent Trolox in FRAP.Salidroside,ligustroflavone,specnuezhenide,oleuropein,oleanolic acid and ursolic acid correspond to S12,S25,S27,S28,S41 and S42,respectively.Chapter 3 Dynamic Change of Four metal elements in Ligustrum lucidum Ait During Fruit and Leaf GrowthThis chapter determinated four kinds of heavy metal including lead(Pb),cadmium(Cd),mercury (Hg)and arsenic(As)contents in Ligustrum lucidum Ait during fruit and leaf growth by Inductively Coupled Plasma Optical Emission Spectrometer.We explored the changes of heavy metal content by microwave digestion instrument with HNO3 being the digestion acid system.The contents of four metal element changed much in different periods,The content of Cd remained stable,As and Ph were changed in into a parabolic,Hg reached the maximum value at the end of October.After validation,the method was proved to be accurate,sensitive and reliable.The content of heavy metals in different periods of Ligustri Lucidi Fructus and Ligustri Lucidi leaves accord with related national standards. |
