Objective To analyze the allelic ratio(AR)of FLT3-ITD in acute myeloid leukemia(AML)patients.To explore the cutoff value of FLT3-ITD AR with maximum clinical impact and compare the clinical features and CR rate between high AR patients and low AR patients.Methods A total of 42 AML patients in our center were retrospectively analyzed from June 2016 to March 2017.Genomic DNA was extracted from patients’ bone marrow sample at diagnosis.We assessed FLT3-ITD mutation in these patients with PCR and sequencing.FLT3-ITD-positive samples were further analyzed for the ITD allelic ratio(ITD-to-mutant-wild type ratio).Furthermore,we analysis the clinical characteristic and CR rate of patients and find a clinical threshold.The statistics was performed by software SPSS 20.0.Results AR of 62 patients varied from 0.12 to 3.55,the median value is 1.06.For the 42 patients,CR rate of higher AR group is 30%,while the low AR ratio group has a CR rate of 75%,(P=0.027)。For the 33 AML(non-APL)patients,higher AR group has a CR rate of 25%,and the low AR ratio group has a CR rate of 68%,(P=0.048).There’s no difference in clinical characters including gender,age,blood cell count and other gene mutations.Conclusions Patients with high FLT3-ITD AR presented with lower CR rate.There’s no difference between higher AR and lower AR patients in clinical features.
