Objective:To determine the association of SATB1 expression with the sensitivity to radiotherapy among rectal cancer patients. Then it can be distinguish different individuals of neoadjuvant therapy beneficiaries from the patients. We can provide the reference to rectal cancer patients with individualized treatment.methods:The clinical data of 78 rectal cancer patients was retrospectively collected for patients who received preoperative radiotherapy(2 Gy/d,x25 d)and meeting into the set of conditions from January 2014 to April 2017 in the second affiliated hospital of nanchang university.Pretreatment tumor biopsies were immunostained for SATB1 expression.Rectal cancer regression grade(RCRG)was used to evaluate the sensitivity of the rectal cancer to preoperative radiotherapy. The correlation of SATB1 expression and sensitivity to radiotherapy was analyzed.Results:SATB1 differentially expressed in rectal cancer tissue with 41 high expression and 37 low expression. There were 36 cases of radiation sensitivity group and 42 cases of radiation non-sensitivity group.Prior to postoperative treatment,SATB1 showed enhancement:4 cases of radiation sensitivity,31 cases of radiation non-sensitivity.SATB1 showed no enhancement:32 cases of radiation sensitivity,11 cases of radiation non-sensitivity.The two groups were statistically different(p<0.05).The expression of SATB1 was associated with rectal tumor invasion lumen diameter、the differentiation 、lymph node metastasis and infiltration depth of rectal cancer.Rectal cancer with higher expression of SATB1 among poorly differentiated 、infiltration depth、 low lumen diameter more than a quarter and presence of regional lymph node metastasis rectal cancers(all P<0.001).Among the forty-one cases SATB1 high patients, two patients showed 1st RCRG,two patients showed 2nd RCRG and thirty-seven patients showed 3rd RCRG.For the thirty-sevencases SATB1 low patients, there were eighteen 1st RCRG,fourteen 2nd RCRG and five 3rd RCRG.There was a significant association between SATB1 expression and sensitivity to radiotherapy(P<0.0001).Multivariate logistic regression analysis showed that the expression level of SATB1(P<0.0001)and the differentiation of rectal cancer(P<0.0001)、infiltration depth(P<0.0001)and lymph node metastasis(P<0.001)were independent predictive factors for the sensitivity of rectal cancer to radiotherapy. Conclusions:Correlation between SATB1 expression and sensitivity to radiotherapy of rectal cancer may exist, which may be helpful in predicting the sensitivity of rectal cancer to preoperative radiotherapy.
