| Objective:To investigate the abnormal expression of Ezrin and PTEN protein in intrahepatic cholangiocarcinoma(ICC)and para-carcinoma tissue,and explore the relationship between Ezrin and PTEN protein and the clinicopathologic significance of those two proteins in the carcinogenesis and development of ICC.Methods:50 cases of ICC tissue(ICC group)and corresponding para-carcinoma tissue(2cm or more to the tumor margin and no tumor cells confirmed by pathology)(control group)were collected from the First Affiliated Hospital of Nanchang University from January 2011 to December 2015.All patients had full clinicopathologic data and didn’t undergo radiotherapy,chemotherapy and other adjuvant therapy before surgery.The expression of Ezrin and PTEN protein in the two groups were detected by using SP immunohistochemistry(IHC),and the experimental clinicopathologic parameters were statistically analyzed by SPSS17.0.Results:1 、 Ezrin was mainly located in cytomembrane and cytoplasm.The positive expression rate of Ezrin was respectively 78.00%(39/50)and 46.00%(23/50)in ICC group and control group;The different expression between the two groups is statistically significance(P<0.05).The expression of Ezrin in ICC group were highly related to tumor size,differentiation grade 、TNM stage,and lymphatic metastasis(P<0.05),but it wasn’t related to age,gender,serum CA19-9 level,hepatitis B virus infection,calculus of intrahepatic duct(P>0.05).2、PTEN was mainly located in cytoplasm.The positive expression rate of PTEN was respectively 27.19%(13/50)and80.95%(35/50)in ICC group and control group;The different expression between the two groups is statistically significance(P<0.05).The expression of Ezrin in ICC group were highly related to differentiation grade、TNM stage,and lymphatic metastasis(P<0.05),but it wasn’t related to age,gender,serum CA19-9 level,hepatitis B virus infection,tumor size,calculus of intrahepaticduct(P>0.05).3、Spearman correlation analysis showed that there was a negative correlation between the expression of Ezrin and PTEN protein in ICC(r=-0.382,P=0.006<0.01).Conclusions:1、Ezrin may promote the invasion and metastasis of ICC.2、PTEN may inhibit the invasion and metastasis of ICC.3、The negative correlation between the expression of Ezrin and PTEN protein in ICC may play an important role in the invasion and metastasis of ICC. |
