Objective:Immunohistochemistry was performed on paraffin sections from patients with colorectal cancer to resarch the expression of YAP and RhoA in cancer tissues and adjacent tissues.Then we investigate the difference of the expression of the two groups in the paraffin sections of colorectal cancer tissues and adjacent tissues,and to analyze the correlation between the two factors and other clinical factors with prognosis.Methods:1.We collect the Paraffin samples and pathological data of 50 patients with colorectal cancer all coming from the First Affiliated Hospital of Nanchang University from October 2010 to December 2011.All patients were treated by postoperative pathological diagnosis of colorectal cancer,the patient received neoadjuvant chemotherapy before operation were excluded.2.Immunohistochemical staining was used to detect the expression of YAP and RhoA protein in paraffin sections of patients with carcinoma and adjacent tissues.3.Data analysis software for SPSS 22.The relationship between the expression level of YAP and RhoA protein and clinical parameters of patients with chi square test,if T<5,then we use Fisher exact probability method to analysis.Kaplan-Meier survival analysis was used to analyze the relationship between the expression of the two proteins and prognosis.Finally,COX risk regression analysis was used to analyze the factors.Results:1.The expression of YAP and RhoA protein in colorectal cancer tissues and adjacent tissues was significantly higher than that in adjacent tissues.The high expression rate of YAP protein in cancer tissues was 52%(26/50),The high expression rate of adjacent tissues was 48%(24/50),X~2=16.327,P<0.001,The high expression rate of Rho A protein in cancer tissues was 60%(30/50),Paracancerous tissue 40%(20/50),X~2=13.3,P<0.001.2.The expression of YAP protein was compared with age,sex,depth of invasion(T grade),number of regional lymph node metastasis(N)and clinical stage of TNM,Results show: The expression of YAP was related to the depth of invasion,The number of regional lymph node metastasis and TNM staging.Irrespective of age and sex.Using the Fisher exact probability method to analysis the statistical data of T1+T2 to contrast that of T3+T4,it shows that P=0.02,meaning the expression of YAP protein was correlated with the depth of tumor invasion.Then we use the same way to analysis the statistical data of group N,P=0.002,meaning the expression of YAP protein was also correlated with the number of regional lymph node metastases.Similarly,using Fisher test,the expression of YAP was correlated with clinical TNM stage,P=0.001.3.We use the same way to analysis the expression of RhoA protein,Results show: The expression of RhoA was correlated with the number of lymph node metastasis and TNM stage,but not with the depth of tumor invasion,age and sex.Using the Fisher exact probability method to analysis the statistical data of group N,P=0.014,meaning the expression of RhoA protein was correlated with the number of regional lymph node metastases.as the same way,we get similar results of the correlation of RhoA and the clinic TNM staging,P=0.022.4.Univariate Kaplan-Meier survival analysis showed that YAP,RhoA protein expression,tumor invasion depth(T)and regional lymph node metastasis(N)were the prognostic factors of rectal cancer,P<0.05,the difference was statistically significant.We brought these above factors into the COX risk regression model.Then we find that the expression level of YAP protein and the number of regional lymph node metastasis(N)were independent prognostic factors for colorectal cancer.Conclusion:1.The expression of YAP and RhoA protein in colorectal cancer patients increased significantly.2.The expression levels of YAP and RhoA were related to the depth of invasion(T),Regional lymph node metastasis(N,No lymph node metastasis was N0,1-3 lymph node metastasis was N1,>3 was N2)and Clinical TNM staging;not related to sex and age.3.YAP protein and regional lymph node metastasis were independent prognostic factors in patients with colorectal cancer.
