Objects:This paper analyzes how As2O3,the main component in arsenic,will influence on the expression of EZH2 mRNA in u937 cell group in Acute Myelogenous Leukemia(AML),thus to provide new methods and ideas for the research on pathogenesis and curing of leukemia from biology functions of EZH2 genes.Methods:Adopt CCK-8 method to detect how different concentrations of As2O3 will inhibit the proliferation of u937 cell line;under the inversed microscope,observe and photograph the morphology of regular cells and u937 cells cultivated under different As2O3 concentrations;use Real-Time PCR method to detect the expression of EZH2 mRNA in u937 cells before and after medication.Results:Cultivated u937 cells with different concentrations of As2O3,which provided different degrees of inhibitions for the proliferation of u937 cell line,and found that the inhabitation rate increased with concentration.Separately chose 25nmol/L,50nmol/L,100nmol/L and 200nmol/L concentrations of-As2O3 in which u937 cell lines were cultivated for 24h,and found that the expression of EZH2 mRNA deceases under different degree.Comparing each experimental group with control group and within each experimental group,the expression differences of EZH2 was found to be statistical(P<0.05).Therefore,As2O3 can enhance the apoptosis of u937 cells,and the degree depends on time and dosage.Conclusion:As2O3 can inhibit the proliferation of u937 cell lines and induce the apoptosis,lowering the expression level of EZH2 mRNA.
