Study Of Serum Pharmacochemistry About Dahuang Fuzi Decoction

Dahuang Fuzi Decoction(DFD),a composite prescription of traditional Chinese medicines,comprising Radix et Rhizoma Rhei,Radix Aconiti Lateralis Praeparata and Radix et Rhizoma Asari,is widely used to treat appendicitis,biliary colic,chronic dysentery,acute ileus and adhesive ileus in clinic,which was originally recorded by Zhang Zhongjing in a publication entitled ’Synopsis of Golden Chamber’(Jin Kui Yao Lue).This formula is complex chemical composition,including anthraquinones,alkaloids and volatile oil.The main task of this topic is to establishing the analysis method of DFD based on serum pharmacochemistry theory.Focus on the vitro and in vivo exchanges of chemical composition of DFD,lay the foundation for the further study on the material basis of DFD to the efficacy of warming interior and dissipating cold.The study comprises five parts:First of all,for the quality evaluation of hurb slices,we collect batches of the three medicinal herb slice samples from different medical institutions nationwide,and determine the content of the index components or the limited component following the way documented in Ch.P.It is no doubt that the medicinal herbs slected in the formula are of good quality.The second part is establishment of a accurate and reliable method for the quantification of the ten different components simultaneously in DFD just by high performance liquid chromatography(HPLC)coupled with electrospray ionization tandem mass spectrometry(ESI-MS)and ultraviolet(UV)detector,to clarify the effect of compatible medicinal herbs on concentration of components in decoction and provide a reference for quality control.The third part is development and validation of the HPLC characteristic spectrum of DFD for the chemical analysis about DFD in vitro,analyzing index components and composition in DFD,determining their relegations and laying methodological foundation for the further stydy in vivio.The fourth part is the establishment of the methods of preparation and pretreatment of serum sample of rats after giving DFD,and to carry out a preliminary serum parmacochemistry’ through analyzing the compounds absorbed into blood based on HPLC characteristic spectrum of DFD.HPLC fingerprints of the serum samples collected after giving DFD and single crude drug are compared,and the constituents absorbed into the serum after DFD administration are analyzed.And further structural analysis of the blood components have been made by HPLC-Q-TOF/MS technology and 22 components into the blood are identified.The last part is the text of the conclusion,with a summary to the research of serum pharmacochemistry of Dahuang Fuzi Decoction and a view of innovations and imperfections in this thesis,and bring forward the next phase of the proposed research prospects.
