Rabbit Nonsustained Ventricular Tachycardia Establishment Of An Animal Model With The Tachycardia Cardiomyopathy And Relation Research

Objective:l.to establish the tachycardiomyopathy animal model of nonsustained ventricular;2.preliminary discuss an the relationship between nonsustained ventricular tachycardia and tachycardiomyopathy;3.preliminary discuss the impact oral levocarnitine injection on tachycardiomyopathy.Methods:The research object and grouping:25 New Zealand rabbits(bought from Hunan People’s Hospital,Institute of clinical medicine),according to the same sex,of similar weight,health condition,all animal after health and epidemic prevention departments inspected.It will be divided into five groups.The control group(a group n=5):Via right anterior vena cava implanted of homemade 5F arterial sheath,but not stimulus;(2)Group bln=5(pacing for 1 weeks):immediate death;(3)Group b2(n=5)pacing for 1 weeks ago with the b1 group 1 weeks after cessation of pacing,pacing and drug intervention(oral levocarnitine injection production:Sigma-tau,100mg/kg);(4)Group b3(n=5)pacing for 1 weeks ago with the b1 group,pacing for 1 weeks after pacing in 2 weeks with Effect of L-carnitine drug;(5)Group b4(n=5)1 weeks after cessation of pacing,pacing and without pharmacological intervention,observation to 2 weeks.Stimulus method:operation process with control group.Using 500ml with heparin injection of 1.25 × 104 units of heparin water sheath prevention of embolism,conventional intramuscular injection of penicillin of 800000 units of infection two times a day.10 coronary sinus electrode implantation of the puncture sheath to the right ventricle(ECG is visible on the premature ventricular contraction and the thumb and index finger touching the electrode with the heart beat can be judged in the right ventricle),connection program stimulation instrument,The perception to the minimum,voltage 3.0±0.5volt;pacing frequency of 400times/min,pacing of right ventricle,30 Bursts per minute,each array continuous 5-6premature ventricular contractions,without experimental factors;Each stimulus is the right ventricular electrode withdrawal,but not withdrawal in vitro,again when stimuli in awake to insert,avoid coronary sinus electrode catheter cardiac cavity on hemodynamic effects.The stimulation process for continuous ECG monitoring(connected the ECG machine,model GE,with self-made needle into animal limbs acquired ECG shape),each time after stimulation with povidone iodine disinfection incision infection prevention,a week after leaving the serum and heart specimens.In the preoperative,postoperative1 weeks,2 weeks after operation to observe four groups of tachycardiomyopathy Animals in the pathogenesis of different period of performance,there is no postoperative infection,Comparison of the level of serum BNP,MMP-9,TIMP-1 concentration,the heart color Doppler sonography of hemodynamic changes and the housing cavity size and ejection fraction size changes,The use of HE staining method,electron microscope observation of extracellular fibers respectively the changes and the period of myocardial ultrastructure morphological changes.Results:after 4 days of pacing group pacing are fatigue,loss of appetite,hair shiny,dull eyes in performance,1cases of postoperative complications such as infection,pericardial effusion.bl、b3、b4group than in the preoperative LVEDd increased significantly(p(0.05),BNP、MMP-9 increased significantly(p(0.05),EF、FS decreased significantly(p<0.05),HE staining showed myocardial structural derangement,myocardial fibrosis,electron microscopy showed mitochondrial swelling,dissolution,or even damage,Z line fuzzy,intercalated disc display is unclear.b2group than in the preoperative had significant change(p(0.05).Conclusion:1.No X-ray,a thoracic vein with electrode mapping procedure to stimulate and successfully establish nonsustained ventricular tachycardia of tachycardiomyopathy animal model,and is simple and easy to operate;Nonsustained ventricular tachycardia≥300array/24hours,each array10-12ventricular extrasystoles,lasting 1 weeks can lead to left ventricular enlargement,reduced ejection fraction;timely termination of nonsustained ventricular tachycardia after 1 weeks after the cardiac function and myocardial structure of partially reversed,but whether can be fully restored,need remain further study;3.No termination of nonsustained ventricular tachycardia may develop irreversible cardiomyopathy;4.Oral levocarnitine on tachycard-iomyopathy has a certain therapeutic effect.
