Objective:To establish rat model of methamphetamine dependence,then the animals were intervened with gastrodin,to detect the expression of neurotrophic receptor in six cerebral regions related to MA dependence,which were frontal cortex(FC),striatum(CPu),nucleus accumbens septi(NAc),hippocampus(HP)and substantia nigra(SN),and ventral tegmental area(VTA).The dependent mechanism of MA and the effectsgastrodin on the neurotoxicity damages induced by MA were investigated.Methods:80 SD rats were divided into two groups,which were control group,2 weeks group and4 weeks group.The rat model of methamphetamine dependence was established by intraperitoneal injection of MA.After confirming the model was established successfully by observing the experiment of conditioned place preference(CPP)and the stereotyped behavior(SB),the animals were injected MA for 2 weeks,4 weeks,respectively,to establish the model of different dependence periods.And then the rats were intervened by different concentration of gastrodin.The methodology of in immunofiuorescence,western blotting and quantity RT-PCR were applied to detect the expression of TrkA.TrkB.TrkC and p75NTR in six regions.Results:1.Compared with the control group,the scores of stereotyped behavior and CPP of four MA dependent groups had significant difference(P<0.05),which means the rat model of methamphetamine dependence was established.2.Expression of TrkA.TrkB.TrkC and p75NTR were different in the different cerebral regions of same MA dependent group.3.The residence time in the medicine box was significantly shortened after the intervention of gastrodin.The stereotyped behavior score was also significantly decreased after the intervention of gastrodin.4.2 weeks group:compared with the control group,the expression of TrkA、TrkB、TrkC and p75NTR in the different cerebral regions after the intervention of gastrodin had obvious rise.Compared with the group dependent on MA,the expression of TrkA、TrkB、TrkC and p75NTR in the different cerebral region after the intervention of gastrodin had obviously rise,there was no obviously different bettwen different concentration.5.4 weeks group:compared with the control group,the expression of TrkA、TrkB、TrkC and p75NTR in the different cerebral region after the intervention of gastrodin had obvious rise.Compared with the group dependent on MA,the expression of TrkA、TrkB、TrkC and p75NTR in the different cerebral region after the intervention of gastrodin had obvious rise,there was no obvious different betwen different concentration.6 There was no obvious different bettwen different time quantum group after the intervention of same concentration gastrodin.Conclusion:1.The rat model of MA dependence was established successfully by using the method of intraperitoneal injection MA,observing the conditioned place preference(CPP)and stereotyped behavior(SB).2.Expression of TrkA、TrkB、TrkC and p75NTR were different in the different cerebral region after intervened by gastrodin without MA,prompt gastrodine on central nervous system has protective effect.3.Expression of TrkA、TrkB、TrkC and p75NTR had significant difference in the different cerebral regions of same MA dependent group,they might have an intimate correlation with the mechanism of MA dependence.4.After intervened by gastrodin,the expression of TrkA、TrkB、TrkC and p75NTR improved in six cerebral regions.The data indicated that gastrodin might have a protective effect on the neurotoxicity induced by MA.
