BackgroundEsophageal cancer(EC)is one of the most common malignancies worldwide.Its incidence ranks the eighth in malignant tumor,and the mortality rate is the sixth leading cause of cancer death.Esophageal cancer patients with 5 years of survival rate is very low,there are only 10-40%.In China esophageal cancer still is one of the high incidence of malignant tumors.After gastric cancer,lung cancer and liver cancer,the mortality rate is the fourth leading cause of cancer death.Currently,the treatments mainly in surgery,radiation and chemotherapy,while the effect has not been satisfied yet,75% of the patients were died within 1 year after diagnosis.So it is very important to find the specific molecular associate with esophageal cancer development.Using high-throughput miRNA chip hybridization technology in the same TNM stage and prognosis of different esophageal tissue samples,we found that the expression of microRNA-197(miR-197)increased significantly in group with poor prognosis.In this research,miR-197 was studied at the molecular level in esophageal cancer cells.The results further help to clarify the role of miR-197 in the occurrence and development of esophageal cancer,and will lay the theoretical and experimental basis for the molecular classification,prognosis judgement,selection of esophageal cancer and its target gene in clinical treatment.ObjectiveTo investigate the influence of abnormal expression of miR-197 on the proliferation,apoptosis and migration of esophageal cancer Eca109 cell.MethodsThe expression of miR-197 in different esophageal cancer cell lines was detected byreal-time fluorescent quantitative PCR(qPCR).The expression of miR-197 in esophageal cancer cell line Eca109 was targeted by miR-197 lentiviral expression vector.Methyl thiazolyl tetrazolium(MTT),plate clone formation assay,flow cytometry,transwell chamber experiment and wound healing method were applied to observe the influence of abnormal expression of miR-197 on the biological characteristics of esophageal cancer Eca109 cell.Results(1)Result of qPCR showed that the expression of miR-197 in esophageal canc er cell line Eca109 was significantly lower than that in HECC(P<0.01);(2)After transfected with lentiviral expression vector,the expression of miRNA-197 in esophageal cancer cell line Eca109 obviously upregulated(P<0.01);(3)MTT and wound healing experiment showed that over-expresssion of miR-197 inhibit esophageal cancer cells Eca109 proliferation(P<0.05);(4)Flow cytometry and qPCR were used to detect the apoptosis rate.The resu lts showed that the over-expression of miR-197 enhance the apoptosis of esophageal cancer cells Eca109(P<0.05);(5)Transwell cell test and scratch test results show that compared with negativ e control group,miR-197 expression can significantly inhibit cell migration(P<0.05).ConclusionThe expression of miR-197 in esophageal cancer cells Eca109 was lower than HECC cell.Up-regulation of miR-197 could inhibit esophageal cancer cells Eca109 proliferation,migration and enhance the apoptosis of esophageal cancer cells Eca109.
