Objective:To compare the expression of Adenosine 2A Receptor in the sperm of normozoospermic and asthenozoospermia men and investigate the correlation of Adenosine 2A Receptor with sperm motility,fertilization rate,embryo quality and pregnancy rate in the cycle of fresh In Vitro Fertilization and Embryo Transfer(IVF-ET).Methods:Firstly,using Real-time PCR and Western Blot analysis to detect the expression of A2AR in human sperm and Using immunofluorescence technique to detect the location of A2AR in sperm.Comparing the content of sperm A2AR between normozoospermic and asthenozoospermia patients.Secondly,the data with IVF-ET from January 2011 to April 2015 were collected and compared the fertilization rate,high quality embryos rate,pregnancy rate between the normozoospermic and asthen-ozoospermia patients for analyzing the relation of ART outcome with asthenozoospermia.Lastly,the sperm with IVF-ET from January 2016 to December 2016 were collected and detected the content of A2AR.According to the content of A2AR,the selected patients were divided into the normal A2AR group(n=59)and the down A2AR group(n=44).Comparison the fertilization rate,high quality embryos rate,biochemi-cal pregnancy rate,pregnancy rate among the two groups of patients.Results:RT-PCR and Western Blot revealed A2AR expressed in human sperm and the content of A2AR declined in asthenospermia patients.Immunofluorescence technique detected the A2AR located in sperm tail.The retrospective analysis between the normal and asthenospermia patients revealed that the fertilization rate and pregnancy rate of the asthenospermia patients were significantly lower than the normal patients(76.84%VS79.65%,59.73%VS67.61%,P<0.05),and the mean age of women and men,retrieved oocytes and high quality embryos rate were without significant difference(P>0.05).The comparative result of the normal A2AR group and the down A2AR group revealed that the fertilization rate of normal A2AR group was significantly higher than the down patients(71.93% VS64.98%,P<0.05),as well as the high quality embryos rate,biochemical pregnancy rate,pregnancy rate(33.72% VS 31.25%,74.00%VS 64.86%,68.00%VS 59.46%),but without significant difference(P>0.05).The mean age of women and men,retrieved oocytes were without significant difference(P>0.05).Conclusion:The A2AR exists in mature human sperm and locates in sperm tail.The A2AR may associates with the asthenospermia,fertilization rate,embryo quality and clinical pregnancy.
