| Hepatitis B is a global public problem,which is threatening human health.At present,the hepatitis B prophylactic vaccine has been proved to be high immunogenicity,protective and effective.The treatment of hepatitis B is to prevent the progression of hepatic fibrosis,liver failure,and hepatocellular carcinoma.There are some anti-hepatitis B virus medicines have applied in clinic,but these drugs are unable to solve the liver injury problems,and drug resistance is also one of difficulties in the treatment of hepatitis B.Therefore,it is urgent to develop a method that can completely cure hepatitis B,and the therapeutic vaccine is one of the most potential therapeutic methods.Researchers have tried a large numbers of vaccine construction methods and immunization strategies to improve the effectiveness of hepatitis B vaccine.Using cytokines as an adjuvant for vaccine is one of the strategies[1].Some studies have confirmed that IL15 can enhance the antigen specific cellular immune response,the specific CTL response and the survival of T memory lymphocytes[2].The recombinant plasmid pVR-S,recombinant virus rMVA-S and recombinant virus rAdv-S,which all expressing the HBsAg gene was selected to research the influence of pVR-IL15 gene adjuvant on the immune response induced by different immunization strategies.The results showed that pVR-IL15 gene adjuvant can enhance the humoral immune response induced by all immunization strategies except in strategy immunized with recombinant virus alone.rAdv-S recombinant virus induced higher humoral immune response than rMVA-S in immunized with recombinant virus alone strategy,once PVR-S and once recombinant virus strategy,and immunized with twice pVR-S and once recombinant virus strategy.The humoral immune effect was induced the most strongly by the strategy of twice pVR-S and once recombinant virus immunization,then it was milder induced by once pVR-S and once recombinant virus strategy,and immunization alone recombinant viral strategies was the weakest.humoral immune response induced by two doses rMVA-S was weaker than the cross immune rAdv-S and rMVA-S strategy.the cellular immune response results showed that pVR-IL15 gene adjuvant can enhance the cellular immune response induced by all immunization strategies in this reacher.There was no difference between the cellular immune level of homologous-vector virus and heterologous-vector virus in immunized twice with recombinant virus strategy;the cellular immune level between immunized with once pVR-S and once recombinant virus strategy and immuned with twice recombinant virus strategy was no difference;immunized twice pVR-S and twice recombinant virus strategy induced cellular immune response level in each immunization strategy was the strongest.In conclusion,pVR-IL15 adjuvant can enhance the humoral and cellular immune response induced by recombinant virus vaccine.The immune effect induced by rAdv-S on humoral immunity is stronger than rMVA-S;humoral immune response to immune twice rMVA-S was weaker than the cross immune with rAdv-S and rMVA-S.there was no difference between immune homologous vector virus and heterologous vector virus in the cellular immune response,the cellular immune response between immunized with once pVR-S and once recombinant virus strategy and immuned with twice recombinant virus strategy also showed no difference.the immune response induced by the combination of the twice recombinant plasmids and the twice recombinant virus was the strongest in both humoral and cellular immunity. |
