Objective:Using the new markup reagent-quantum dots(QDs)in cervical squamous carcinoma tissue for the detection of different protein,and to assess protein expression and significance in cervical squamous cell carcinomas.Methods: In 30 cases Normal Cervix(NC)、 30 cases Low grade squamous intraepithelial lesions cervix(LSIL)、 30 cases High grade squamous intraepithelial lesions cervix(HSIL)and 60 cases Squamous carcinoma of the cervix(SCC),the use of QDs labeled streptavidin complexes(QDs-SA)with biotin Hua Er Ig G resistance combined with the characteristics of immunofluorescence histochemical detection EZRIN protein and FAK in the protein expression.Using immunohistochemical SP method to detect the expression of Ezrin and FAK in cervical squamous carcinoma tissues.Results:1.Colored SP method to observe Ezrin,FAK protein in cervical cell membrane and cytoplasm,tan,and in 60 cases of cervical squamous cancer cells positive expression rate of 75%,80% respectively,Compared with the method of QDs-IHC results,the difference had statistical significance(P < 0.05);2.QDs-IHC method to observe Ezrin,FAK protein was observed respectively located in cervical cell membrane and cytoplasm,accurate positioning,and positive signals were green,orange red light.3.The positive expression rate of Ezrin and FAK protein in 60 cases of cervical squamous carcinoma tissues were 96.66% and 95.00% respectively,compared with the HSIL group was statistically significant difference(P < 0.05).The positive expression rate of Ezrin and FAK protein in 30 cases of HSIL group were 83.33% and80.00% respectively,compared with the LSIL group was statistically significant difference(P < 0.05).The positive expression rate of Ezrin and FAK protein in 30 cases of LSIL group were 60.00% and 53.33% respectively,compared with the normal cervix group was no statistically significant difference(P >0.05).Conclusion:The positive expression of Ezrin and FAK in cervical squamous carcinoma occurred in the process of development and has a certain clinical significance,is worth further study.QDs-IHC method is better than the SP method for the simultaneous detection of multiple indexes.
