Purpose : To determine the clinical significance of TFF2 expression and Helicobacter pylori infection in patients with gastric cancer. Methods: TFF2 and Helicobacter pylori in gastric specimen was detected by immunohistochemistry. The correlations of TFF2 expression and Helicobacter pylori infection with clinicopathological characteristrics of patients were analysed. The Kaplan-Meier survival analysis was used to construct survival curves, and then used the log rank test to calculate the P value.Assessed the prognostic value in patients with different variables by the Cox proportional hazards regression model analysis.Results:1.The clinicopathologic characteristics of gastric cancer patientsA total of 82 patients were included in the study.55 patients were men,and 27 patients were women in this study. The ratio of male to female was 2.0:1.Their age ranged 37-82 years old.The average age was 59.2±11.5 years old.The TNM staging system:14 patients with Stage I,19 patients with Stage II,42 patients with Stage III,7 patients with Stage IV.The depth of invasion: T3 were the most commom classification.10 patients with T1,8 patients with T2,60 patients with T3,4 patients withT4.There were 50 patients with lymph nodes metastasis, and 32 patients without lymph nodes metastasis. There were 55 patients poorly-moderately differentiated, and 27 patients were well differentiated.Tumor size:47 patients with diameter ≤4cm,and 35 patients with diameter>4cm.2.The relationship between TFF2 expression,HP infection and clinicopathological characteristics of gastric cancerWe examined that TFF2 mainly stained in cytoplasm. In 30 cases of gastric normal tissues, the expression rate of TFF2 was 83.3%(25/30)。There were 45(54.9%)patients of gastric cancer expressed TFF2.The TFF2 expression in gastric cancer specimen was significantly lower than gastric normal tissues(P<0.05).The expression of TFF2 was significant difference between poorly-moderately differentiated and well differentiated tumor(P<0.05).There were 50(61.0%)patients with Helicobacter pylori positive infection. The status of Helicobacter pylori infection showed significant difference with depth of invasion,TNM stage(P<0.05).3.Prognosis analysisUnivariate analysis showed that the age,TNM stage, tumor diameter, tumor differentiation, depth of invasion, lymph node metastasis, the expression of TFF2 and Helicobacter pylori infection status were significantly correlated with the survival of patients with gastric cancer.Multivariate analysis showed that lymph node metastasis, tumor diameter, the expression of TFF2 and Helicobacter pylori infection status were independent prognostic indicators in patients with gastric cancer. Conclusion: The expression of TFF2 in gastric cancer tissues was significantly lower than that in normal gastric tissues. The expression of TFF2 was related to tumor differentiation.Helicobacter pylori infection was related to the depth of tumor invasion and TNM stage. The expression of TFF2 and Helicobacter pylori infection status may be independent prognostic factors in patients with gastric cancer.Detection of TFF2 and Helicobacter pylori infection status could be helpful for prognosis evaluation in gastric cancer. There was a negative correlation between TFF2 expression in gastric cancer and Helicobacter pylori infection, suggesting that there may be a interaction mechanisms between the two factors affecting the occurrence and development of gastric cancer.
