Purpose:To investigate the expression of CD4~+CD25~+Treg cells and CD4~+Treg cells in bladder urothelial carcinoma,and to explore the correlation between them and the clinical indicators of bladder urothelial carcinoma.Methods: Department of Urology,Affiliated Hospital of North Sichuan Medical College from March 2012 to May 2015,50 cases of bladder urothelial carcinoma were obtained by plasma resection of bladder tumor.The pathological type and pathological grade were confirmed by the pathology department of Affiliated Hospital of North Sichuan Medical College;normal bladder tissue in 20 cases,data on normal bladder neck mucosa in patients with prostate plasma resection.The expression of CD4~+CD25~+Treg cells and CD4~+Treg cells in bladder urothelial carcinoma and normal bladder tissues were detected by immunohistochemical ElivisionTM plus / HRP method.To investigate the correlation between CD4~+CD25~+Treg cells and CD4~+Treg cells of clinical indicators in bladder urothelial carcinoma.Result:1.CD4~+CD25~+Treg cells were all expressed in bladder urothelial carcinoma,were not expressed in normal bladder tissues.2.The expression of CD4~+CD25~+Treg cells in bladder urothelial carcinoma was not significantly different from that in patients with age,sex,tumor size and tumor number(P>0.05).The expression of CD4~+CD25~+Treg cells in bladder urothelial carcinoma was significantly correlated with pathological grade(r=0.917),the difference was statistically significant(P <0.05),and there was a significant positive correlation with lymph node metastasis(r=0.723),the difference was statistically significant(P<0.05),and there was a significant positive correlation with recurrence(r=0.670),the difference was statistically significant(P<0.05).3.CD4~+Treg cells were all expressed in bladder urothelial carcinoma,were not expressed in normal bladder tissues.4.The expression of CD4~+Treg cells in bladder urothelial carcinoma was not significantly different from that in patients with age,sex,tumor size and tumor number(P>0.05).The expression of CD4~+Treg cells in bladder urothelial carcinoma was significantly correlated with pathological grade(r= 0.778),the difference was statistically significant(P<0.05),and there was a significant positive correlation with lymph node metastasis(r=0.852),the difference was statistically significant(P<0.05),and there was a significant positive correlation with recurrence(r=0.806),the difference was statistically significant(P<0.05).5.CD4~+CD25~+Treg cells and CD4~+Treg cells in bladder urothelial carcinoma were significantly correlated(r=0.848),the difference was statistically significant(P<0.05).Conclusion:1.The expression of CD4~+CD25~+Treg cells was significantly correlated with the pathological grade,lymph node metastasis and recurrence of bladder urothelial carcinoma.Increased expression of CD4~+CD25~+Treg cells in bladder urothelial carcinoma may help determine the recurrence and prognosis of bladder urothelial carcinoma.CD4~+CD25~+Treg cells may promote the development of bladder urothelial carcinoma.2.The expression of CD4~+Treg cells was significantly correlated with pathological grade,lymph node metastasis and recurrence of bladder urothelial carcinoma.CD4~+Treg cells may play a role in the development of bladder urothelial carcinoma.CD4~+CD25~+Treg cells and CD4~+Treg cells may play a synergistic role in the development and progression of bladder urothelial carcinoma,which may be a potential target for bladder cancer immunotherapy.
