Objective: To investigate the expression and clinical significance of pigment epithelium derived factor(PEDF)in esophageal squamous cell carcinoma(ESCC)and to learn the correlation between the expression of PEDF and microvessel density(MVD)in ESCC.Method: From February 2016 to October 2016 in Nanchong Central Hospital of Sichuan province and North Sichuan Medical college Affiliated Hospital cardiothoracic surgeons,selecting surgical patients with esophageal squamous cell carcinoma and normal esophageal tissues in 64 cases,all patients were not receiving preoperative radiotherapy,chemotherapy and other anticancer drugs.Application of real-time fluorescence quantitative PCR(real-time quantitative PCR,RT-PCR)expression of PEDF around m RNA was detected in 64 cases of ESCC tissue and normal esophageal tissue in the same patient,immunohistochemistry was used to detect the expression of MVD and expression of PEDF in cancer tissue around 64 cases of esophageal squamous cell carcinoma and normal esophageal tissue in the same patient in.Result: The real-time PCR results showed: the expression of PEDF in tissue samples of 64 experimental m RNA were different in intensity,the relative quantitative value of PEDF m RNA in ESCC was 0.77,significantly lower than the surrounding normal esophageal tissues paired(1.04),the difference was statistically significant(P< 0.05).Immunohistochemical detection results showed that:(1)the expression of PEDF protein in 64 cases of ESCC tissues,29 cases of PEDF protein expression was positive and 35 cases of PEDF protein negative expression,the positive expression rate of 45.31%,while 43 cases of PEDF protein was expression in normal esophageal tissues,21 cases of PEDF protein negative expression,the positive expression rate of 67.19%,there is a significant difference between the two groups(P< 0.05).The expression of PEDF protein in the age and sex of the patients,there was no obvious correlation in ESCC(P>0.05),but was correlated with clinical stage,histological differentiation and lymph node metastasis(P<0.05),and the lower the degree of differentiation of tumor tissue,the later stage or lymph node metastasis,the lower the express of PEDF.(2)The expression of PEDF protein and MVD in ESCC: The mean MVD value of 29 cases of ESCC with positive expression of PEDF was 38.3793,while the mean value of MVD in ESCC with negative expression of PEDF was 43.4857 in all cases,and the difference was statistically significant(P<0.05).Conclusion: The expression of PEDF in ESCC was decreased,and the decrease of expression may have some reference value for the prognosis of ESCC.PEDF selective anti-angiogenesis effect and make it become a potential molecular tumor treatment,but its effectiveness,safety,route of Administration(including vector suitable)more studies are required to confirm.
