Hemoglobin is one kind of proteins in human body which is responsible for oxygen transportation.It is in high oxygen affinity when oxygen content is high,on the contrary,it releases oxygen.The concentration of hemoglobin is an important indicator of blood loss anemia,genetic anemia and some other diseases,such as congenital heart disease and obstructive pulmonary emphysema.However,most of current hemoglobin sensors are based on optical detection principle.They are unstable and vulnerable to environmental interference.The goal of this study is to construct a low cost and disposable hemoglobin biosensor based on screen-printed carbon electrode(SPCE)and electrochemical method.Much work has been done to integrate the processes of hemolysis and detection to make the sensor free of extra blood pretreatment.First of all,a prototype SPCE-based hemoglobin biosensor was fabricated with an extra hemolysing step.Then the sensor was optimized in various aspects concerning the choice and concentration of hemolysin,the time for hemolysis,the amount of electron transfer agent and working conditions.The characters of the optimized hemoglobin biosensor were tested,including sensitivity,linearity,repeatability and anti-interference.Experimental results showed that the hemoglobin biosensor with a sensitivity of 0.033 μA/(g/L)in the linear range from 20 g/L to 230 g/L.The RSD values were less than 4.59%and the ability of anti-interference is strong.In clinical trials,the correlation with the standard hospital instrument was 0.9536.Furthermore,a hemoglobin biosensor combining the two procedures of hemolysis and detection was constructed.Hemolysin was selected suited for the further detection process and the hemolysin distribution was designed.The hemoglobin biosensor system based on microfluidic circuit and screen-printed electrodes was designed,fabricated and tested.The final hemoglobin biosensor system showed a sensitivity of 0.041 μA/(g/L)in the linear range from 70 g/L to 230 g/L,as well as a good consistency.
