Expression And Its Clinical Significance Of CRT And BAP31 In Non-small Cell Lung Cancer

Objective: By detecting the expressions of calreticulin(CRT)and B cell receptor associated protein 31(BAP31)in non-small cell lung cancer(NSCLC)tissues,paracancerous tissues,and normal lung tissues and to analyze the relationship between the expressions of these proteins and the age,sex,smoking status,pathological type,lymph node metastasis and the clinical stage of NSCLC patients,and then to further explore the role of CRT and BAP31 in the occurrence,development,invasion and metastasis of NSCLC and to determine the synergistic or antagonistic action of these two proteins in the occurrence and development of NSCLC through the correlation analysis of the two proteins in NSCLC.Methods:Sixty NSCLC samples were selected from patients who underwent thoracic surgical resection and were pathologically diagnosed as NSCLC in the Affiliated Hospital of Southwest Medical University from Jan.2015 to Dec.2015,including 30 cases of squamous cell carcinoma and 30 cases of adenocarcinoma.30 corresponding paracancerous tissues(at least 5cm from the margin of the tumor)were randomly selected,including 17 cases of squamous cell carcinoma and 13 cases of adenocarcinoma.All the samples were used as test objects.All the patients included had complete clinical diagnosis and treatment information and those with other lung diseases and serious heart,liver,kidney,and metabolic diseases were excluded.No patients were treated with radiotherapy,chemotherapy,molecular targeted therapy and other treatments for malignant tumor before the test samples were collected;in addition,30 normal lung tissue samples were selected as the control group.Expression of CRT and BAP31 in NSCLC cancer tissues,paracancerous tissues,and normal tissues were determined by immunohistochemical method,the relation between the expression of these two proteins in NSCLC tissues and the age,sex,smoking status,pathological type,lymph node metastasis,and clinical stage were analyzed,respectively,and the correlation between the expressions of the two proteins in NSCLC was further determined.Results: 1.Positive expression rate of CRTin NSCLC was 88.33%,which was significantly higher than that of the normal lung tissues and the paracancerous lung tissues(6.67% and 13.33%,respectively),and the difference was statistically significant(P<0.01).However,the difference in the CRT expression rate between the normal lung tissues and the paracancerous tissues was not statistically significant(P>0.05).The positive rate of CRT expression in NSCLC patients was not significantly related to the sex,age,smoking status and pathological type,but was correlated with the metastasis and clinical stage of the disease.2.Positive expression rate of BAP31 in NSCLC was 83.33%,significantly higher than that of the normal lung tissues and the paracancerous lung tissues(3.33% and 6.67%,respectively),and the difference was of statistical significance(P<0.01).No statistical difference was noted in the positive expression rate of BAP31 between the normal lung tissues and the paracancerous tissues(P>0.05).The positive expression of BAP31 was not correlated with sex,age,smoking status and pathologic type,and it was not correlated with lymph node metastasis and different TNM staging 3.No significant correlation between the expressions of CRT and BAP31 was found in NSCLC.Conclusion: 1.CRT is highly expressed in NSCLC,and its positive expression is correlated to lymph node metastasis and clinical stage of the disease.CRT may be involved in the occurrence and development of NSCLC.2.BAP31 is highly expressed in NSCLC and and its positive expression was not significantly correlated with age,sex,pathological type,lymph node metastasis and TNM stage.3.There is no significant correlation between the expressions of CRT and BAP31 in NSCLC tissue,which needs to be further confirmed in a large scale study.
