OBJECTIVE:To investigate the role of LPA2 receptors in PC 12 cell injury induced by hypoxia/reoxygenation and the possible mechanismMETHODS:PC 12 cells hypoxia/reoxygenation injury model was established by cell oxygen deprivation(OGD).PC 12 cells were divided into 6 groups:normal group,hypoxia 3h,6h,9h,12h,15h group.After hypoxia,the cells were cultured in normal incubator for 24 hours,MTT cell viability.PC 12 cells were divided into 5 groups:normal group,glucose deprivation(OGD)group,glucose oxygen deprivation +solvent control group(OGD + DMSO)group;glucose deprivation + LPA2 agonist(OGD + Dodecylphosphate)group,LPA2 receptor(OGD + H2L5186303),the survival rate of MTT cells and the expression of LPA2 receptor and p-akt protein were detected by Western Blotting.Results:Cell viability was significantly lower than that in the normal group(P<0.05),and the cell viability was decreased by 50%at 12h after hypoxia.(P<0.05),and the cell viability decreased(P<0.05).There was no significant difference in the solvent control group compared with the glucose-oxygen deprivation group.Compared with the aerosol group(P<0.05).The expression of LPA2 receptor in LPA2 receptor group was significantly lower than that in glucose deprivation group(P<0.05),and the expression of LPA2 receptor was significantly increased in deprivation group(P<0.05).Conclusion:The survival rate of cells was significantly lower than that of the normal group(P<0.05).The survival rate of hypoxia 12h cells decreased by 50%,which was the most suitable for reperfusion injury.Compared with the normal group,the cell viability and LPA2 receptor expression were decreased in the glucose-deprivation group.The expression of LPA2 receptor was increased and the cell viability increased with the addition of agonist.The above results can confirm that LPA2 receptors play a protective role in brain glucose deprivation injury...
