| Objective:To investigate the IL-4 and IL-13 gene polymorphism and China Xinjiang Uygur children bronchial asthma(asthma)and the relationship between the phenotype of IgE(serum total IgE)on the level of understanding,and two sites have a synergistic effect on asthma.Methods:a total of 38 asthmatic children and 30 healthy children,using polymerase chain reaction(PCR)and direct sequencing of IL-13(Arg130Gln)analysis of the promoter region of-590C/T gene polymorphism and IL-4 gene promoter,by enzyme-linked immunosorbent assay(enzyme-linked immunosorbent,assay,ELISA)were detected in two groups and asthma related indexes such as serum total IgE,IL-4,IL-13 and the level of allergens,monitoring of lung function indexes such as airway resistance,FEV1,FEV1/Pred%,and the two groups were compared and analyzed the relevant indicators,and analysis of IL-13(Arg130Gln)and IL-4 gene promoter region of-590C/T gene has no synergistic effect on the pathogenesis of childhood asthma.Results:1.the T allele frequencies of IL-4C-590 T gene in asthma group and control group were significantly higher than those in control group,the two groups were statistically significant.(45.9%,10.3%,OR=8.91;63.5%,36.2%,OR=3.07,P<0.05).2.there was no significant difference in genotype and allele frequency distribution of IL-13 gene between and Arg130 Gln in asthma group and control group.(54.1%,17.2%,OR=6.29;67.6%,39.7%,OR=3.17,P<0.05).3The serum IgE level of different genotypes in 3 asthma group were: the two groups showed the asthma group was significantly higher than the control group.(Z:-2.128,-2.050,-2.700;-3.766,-3.799,-3.397,P<0.05).4.the risk of asthma inpatients with IL-4-590C/T TT genotype was significantly higher than that with only a single L-4-590C/T TT genotype,(OR=6.00,P=0.046;OR=4.50,P=0.033;OR=22,P=0.005).Conclusion: the-589C/T polymorphism of IL-4 gene and Arg130 Gln is associated with the susceptibility to asthma and the increase of serum IgE in Xinjiang Uygur population.The T allele of IL-4 locus-590C/T and IL-13(Arg130Gln)locus A allele had synergistic effect. |
