| Objective:Established the methods of Thin Layer Chromatography(TLC)identification and High Performance Liquid Chromatography(HPLC)determination for the Traditional Chinese Medicine(TCM)Citri Reticulatae Semen and Citri Reticulatae Retinervus to improve these current quality standards.Method:1.The frist chapter: Three Limonoids,namely Limonin,Nomolin and Obacunone,were used as evaluation indexes to optimize preparation conditions of the test solution and TLC conditions.Then,the TLC method for the determination of Limonin,Nomolin and Obacunone were established.The preparation conditions of the test solution and the conditions of HPLC were optimized to establish the determination method of Limonin,Nomolin and Obacunone content.2.The second chapter: Using Hesperidin as the evaluation index,optimizing the preparation conditions of the test solution and the TLC conditions,Then,the of TLC identification method for Hesperidin were established.The preparation conditions of the test solution and the conditions of HPLC were optimized to establish the determination method of Hesperidin content.3.The third chapter: Reviewed the literatures on the herbal evolution,chemical composition,pharmacological action and quality control,summarized the research overview of the TCM C.Reticulatae Semen.4.The fourth chapter: Reviewed the literatures on the herbal evolution,chemical composition,clinical application and quality control,summarized the research overview of the TCM C.Reticulatae Retinervus.5.Chapter 5: Reviewed the literatures on the herbal textual research,chemical composition and quality control,summarized the research overview of the TCM Citri Reticulatae Pericarpium and discussed on the quality.Result:1.In methodology study,the indexes of C.Reticulatae Semen detection method meet the analysis requirements,and the TLC identification method for Limonin,Nomolin and Obacunone is simple and feasible.Seventeen batches of amples were identified by thin layer chromatography,and the results showed that Limonin,Nomolin and Obacunone could be detected.2.In methodology study,the indexes of C.Reticulatae Semen determination method meet the analysis requirements,and the HPLC determination method for Limonin,Nomolin and Obacunone has good accuracy,precision,reproducibility.Seventeen batches of amples were identified by HPLC,and the results showed that the contents of Limonin,Nomolin and Obacunone were not less than 0.58%,0.36% and 0.05%,respectively.3.In methodology study,the indexes of C.Reticulatae Retinervus detection method meet the analysis requirements,and the TLC identification method for Hesperidin is simple and feasible.Thirteen batches of amples were identified by thin layer chromatography,and the results showed that Hesperidin could be detected.4.In methodology study,the indexes of C.Reticulatae Retinervus determination method meet the analysis requirements,and the HPLC determination method for Hesperidin has good accuracy,precision,reproducibility.Thirteen batches of amples were identified by HPLC,and the results showed that the contents of Hesperidin were not less than 3.34%,respectively.5.Study on the status of C.Reticulatae Semen,C.Reticulatae Retinervus,C.Reticulatae Pericarpium for review,analysis and discussion of three medicinal materials of chemical composition,quality standard research problems,provide a theoretical basis for its further research and development. |
