Objective:In this study,the efficacy and safety of different doses donepezil in the treatment of vascular dementia were evaluated by meta-analysis of the randomized controlled trials of cholinesterase inhibitor donepezil in the treatment of vascular dementia,and provided effective evidence for clinical application.Methods:A randomized controlled trial of donepezil in the treatment of vascular dementia was conducted in a database of Chinese academic journals(CNKI)database,Vipu Chinese database,Wanfang database,Pubmed,Cochrane Library,Embase and other databases.Collect literature according to inclusion criteria.669 articles were screened,and eight eligible literatures were extracted,and the data were extracted and analyzed by Revman5.3 software.The efficacy and safety of different doses donepezil on vascular dementia were evaluated.Results:Donepezil 5mg / d group and donepezil 10 mg / d group than the control group in the treatment of vascular dementia have a significant effect,can significantly reduce the simple mental state examination scale(MMSE)score,and no dose-related.The results were statistically analyzed for donepezil 5mg / d and control group: I2 = 21%,WMD = 0.74,95% CI(0.44,1.05);donepezil 10 mg / d and control group: I2 = 0%,WMD = 1.10,95 % CI(0.58,1.62);donepezil 10 mg / d group and donepezil 5 mg / d group: I2 = 0%,WMD = 0.29,95% CI(-0.05,0.64).Conclusion:According to the results of this systematic review,donepezil has a significant improvement in patients with vascular dementia,and no significant dose-related.
