Research purposes:Cancer as a worldwide public health problem,is the second leading cause of human death,wide scope of its incidence,high mortality,serious threat to human health.Chemotherapy is an important method to the non-operative treatment of cancer,but there is high frequency,low utilization rate of drug injection,drug metabolism,side effects et al,its treatment effect is not ideal.Temperature sensitive hydrogel as drug carrier has good biodegradable properties and histocompatibility,applied in local treatment of tumor with increasing local drug concentration,sustained release(longer),dosing frequency is low,the body’s advantages of low toxicity,as drug carrier has been widely used in the research of local treatment of tumor.Drug combination therapy is an effective way to enhance the efficacy of treatment,the current commonly used joint / chemotherapy treatments including surgery,chemotherapy / radiotherapy and chemotherapy / immunotherapy as well as a variety of combination chemotherapy drugs can significantly improve the therapeutic effect.Poly(gamma-ethyl-L-glutamate)(PEG-PELG)hydrogel is a new kind of temperature sensitive hydrogels,can quickly gel in the body,and as an injectable materials for drug carriers.The purpose of this study was to explore the hydrogel material Shared drive histones to acetylation enzyme inhibitors valproate(VPA)and anti-tumor after effective antitumor effect in vitro,provide the basis for further research and clinical application.The details are as follows:The second block polymers PEG-PELG was prepared via ring-opening copolymerization and properties characterization are carried out to determine the degree of polymerization and the molecular weight,PEG2K-PELG15 concrete structure is obtained,its temperature is decreased with the increasing of the concentration,combination of valproic acid sodium and cisplatin cytotoxicity experiments show that after the two drug combination can improve the anti-tumor effect of A549 / CDDP cells,induce apoptosis and cell cycle arrest,co-loaded VPA and CDDP by physical mixing method,cell toxicity experiments show co-loaded the drugs can improve the resistant cells A549 / CDDP antitumor effect,show PEG2K-PELG15 as drug carrier after co-loaded VPA and CDDP can improve the therapeutic effect of A549 / CDDP,and reduce the A549 / CDDP drug resistance,indicting a promising application in lung cancer treatment.In conclusion,the temperature sensitive hydrogels PEG-PELG has good biocompatibility,biodegradable sex and drugs slow-release performance,after take medicine in local tumor therapy has a good application prospect.Valproic acid sodium as histone acetylation enzyme inhibitors and combined with effective cooperative anti-tumor ability,the tumor in vitro explore experiment has the potential research value.
