Objective:The experiment detected the concentration of ANGPTL4 and HMGB-1 in proliferative diabetic retinopathy(PDR)and idiopathic macular hole patients of serum and vitreous liquid,to analysis the differences between them,and also to investigate the concentration difference of ANGPTL4 and HMGB-1 respectively,to provide new ideas for the pathogenesis of PDR.Methods:We had selected 27 patients,16 PDR patients(9 males,7 females)and 11 idiopathic macular hole patients(4 males,7 females).Record the basic information of patients on admission:blood pressure,triglyceride(TG),low density lipoprotein(LDL),fasting blood glucose(FGB),high density lipoprotein(HDL).PDR patients were required to detect blood glucose two hours after meals.In the morning all patients were collected venous blood 3ml from elbow,centrifuged at 4℃ with 3000r/min in 10 min,collected supernatant,and stored in-80℃ super low temperature freezer for the alternate test.PDR patients’ FGB need below 7.8mmol/L,and the blood glucose 2 hours after meals need below 10.0mmol/L.PDR patients were injected anti-VEGF drugs in one week before surgery.Before surgery we collected the central vitreous body liquid 0.3-0.5mL,then centrifuged at 4℃ with 3000r/min in 5min,collected supernatant,and stored in-80℃ super low temperature freezer for the alternate test.Using ELISA to detect the two group’s concentration values of the ANGPTL4 and HMGB-1 in the serum and vitreous.Statistical analysis all the data by using SPSS 21.0 statistical software system.Using t test of two independent samples to express all the normal distribution data.Using Pearson correlation analysis to express the correlation between the two samples.And P<0.05 indicated as significant diference.Results:Differences of age and gender between the control group and PDR groups was not statistically significant(P=0.688,P>0.05)。Differences of ANGPTL4、HMGB-1 in venous between the two groups was statistically significant(P=0.040,0.043)。Differences of ANGPTL4、HMGB-1 in vitreous body liquid between the two group was not statistically significant(P=0.076,0.539).In the serum,there was highly positive correlation between ANGPTL4 and HMGB-1(r=0.753).ANGPTL4 and HMGB-1 showed moderate negative correlation in vitreous(r=-0.515).ANGPTL4 and LDL showed moderate positive correlation in vitreous(r=0.462).ANGPTL4 and cholesterin showed moderate positive correlation in vitreous(r=0.441).Conclusion:1.ANGPTL4 and HMGB-1 all promoted the angiogenesis process of diabetic retinopathy,and there was a possible interaction or the relationship between the ANGPTL4 and HMGB-1.2.Anti-VEGF drugs maybe inhibit the function of HMGB-1 and ANGPTL4.And the inhibitory effect of anti-VEGF on HMGB-1 was far greater than on the ANGPTL4.3.There was a pisitive collection between LDL、cholesterol and ANGPTL4.This suggesting that ANGPTL4 is involved in lipid metabolism in vivo.
